摘 要:目的采用体内和体外试验,研究常用食品添加剂脱氢乙酸钠(DHA-S)的急性毒性效应。方法(1)昆明种小鼠单次经口灌胃4640、2150、1000和464 mg/kg·bw剂量的DHA-S,观察中毒症状和死亡情况,确定半数致死量(LD_(50))。(2)秀丽隐杆线虫(野生型N2)L4期幼虫,分别用0~100 mg/ml DHA-S染毒24 h,计数各剂量组线虫死亡率(%)。(3)体外培养的中国仓鼠肺细胞(CHL),分别用0~5.0 mg/ml DHA-S染毒24 h,计算各剂量组细胞相对增殖率(%)。结果(1)雌性、雄性小鼠急性经口LD_(50)分别为1470和2000 mg/kg·bw,均属低毒级。(2)线虫的LC_(50)为1.53 mg/ml时,具有毒性。(3)细胞LC_(50)为1.78 mg/ml,毒性分级为3级,属中等毒性。结论不同剂量DHA-S急性染毒可对试验动物、模式生物及哺乳动物细胞产生明显的毒性效应。Objective The acute toxic effects of sodium dehydroacetate(DHA-S),a common food additive,were studied in vivo and in vitro.Method(1)Kun-ming mice were given a single oral gavage of 4640,2150,1000,464 mg/kg·bw of DHA-S,and the poisoning symptoms and death were observed to determine the median lethal dose(LD_(50)).(2)The L4 larvae of Caenorhabditis elegans(wild type N2)were exposed to 0~100 mg/ml DHA-S for 24 hours respectively,and the mortality of nematodes in each dose group was counted(%).(3)Chinese hamster lung cells(CHL)cultured in vitro were exposed to 0~5.0 mg/ml DHA-S for 24 hours respectively,and the relative proliferation rate(%)of cells in each dose group was calculated.Results(1)The acute oral LD_(50) of female and male mice were 1470 mg/kg·bw and 2000 mg/kg·bw,respectively,which were of low toxicity.(2)The LC_(50) of nematode was 1.53 mg/ml,which was toxic.(3)The LC_(50) of CHL cells was 1.78 mg/ml,and the toxicity grade was grade 3,belonging to medium toxicity.Conclusion Acute exposure to different doses of DHA-S can produce significant toxic effects on experimental animals,model organisms and mammalian cells.
