摘 要:目的探究番茄红素(LYC)对血管性痴呆(VaD)大鼠认知功能及海马区凋亡相关蛋白caspase9、caspase3表达的影响。方法选用SD鼠随机分为假手术组(Sham组)、番茄红素干预组(LYC组)和血管性痴呆模型组(VaD组)。采用永久性双侧颈总动脉结扎法(2VO)制备VaD模型,灌胃给予LYC10mg·kg·d,共6w。Morris水迷宫实验评价各组大鼠的空间学习及记忆功能,Western blot检测海马区caspase9、caspase3的表达水平。结果VaD组与Sham组相比,VaD组大鼠第1~4天的逃避潜伏期明显延长(P<0.05),穿越平台次数明显减少(P<0.05),海马区caspase9、caspase3表达水平显著增高(P<0.05)。LYC干预后,LYC组与VaD组相比,LYC组大鼠第1~4天的逃避潜伏期明显缩短(P<0.05),穿越平台次数明显增多(P<0.05),空间学习及记忆能力提高,海马区caspase9、caspase3表达水平明显下降(P<0.05)。结论番茄红素可改善VaD大鼠认知功能并降低海马区凋亡相关蛋白caspase9、caspase3的表达。本研究为LYC用于治疗VaD提供了实验依据。Objective To investigate the effects of lycopene(LYC)on cognitive function and the expression of apoptosis-related proteins caspase9,caspase3 in hippocampus of rats with vascular dementia(VaD).Methods SD rats were randomly divided into three groups:the Sham group,the LYC group and the VaD group.VaD model was prepared by 2-vessel occlusion(2VO),and LYC was administrated intragastrically at dose of 10 mg·kg·dfor 6 weeks.The spatial learning and memory function of rats were evaluated by Morris water maze test,and the expression of caspase 9 and caspase3 in hippocampus were quantitatively detected by western blot.Results Compared with Sham group,the learning and memory ability of rats in VaD group decreased,and the expression of caspase9 and caspase3 in hippocampus increased.Compared with the VaD group,learning and memory ability of rats in LYC group increased,and the expression of caspase9 and caspase3 in hippocampus decreased.Conclusion LYC can improve cognitive function and reduce the expression of apoptosis related proteins caspase9、caspase3 in hippocampus of VaD rats.This study provides a theoretical basis for the application of LYC in the treatment of VaD.
