摘 要:该文从开口箭根茎部位提取分离开口箭多糖TCP-C1-1,确定其结构并对其生物活性进行研究.实验采用水提醇沉法从开口箭根茎部分离提取多糖,通过Sevage法对80%醇沉部位TCP-C进行纯化,采用DEAE-52纤维素离子交换树脂和葡聚糖凝胶Sephadex G-200从TCP-C中进行分离纯化得到多糖TCP-C1-1,采用高效凝胶色谱,TLC薄层层析,红外光谱核磁共振氢谱碳谱等技术对多糖TCP-C1-1结构进行分析.结果分析,从开口箭中分离纯化得到TCP-C1-1多糖为果糖组成的均一多糖,连接方式其为β-(2→1)连接,相对分子质量为1.52×104.TCP-C1-1具有良好的生物活性,在浓度为2 mg·mL^(-1)时,其自由基清除率可以达到63.3%.在浓度为6.5 mg·mL^(-1)时,其对α-糖苷酶的抑制率可以达到78.6%.在浓度为1.5μg·mL^(-1)时,其可以明显促进小鼠单核巨噬细胞RAW 264.7的增殖(p<0.05),并能明显抑制LPS诱导小鼠单核巨噬细胞RAW 264.7细胞释放出的NO.TCP-C1-1为首次从开口箭中分离得到的天然来源的菊粉类果聚糖,为初步研究为开口箭多糖成分的开发提供了一定参考.The polysaccharide TCP-C1-1 was extracted and isolated from the rhizome of Tupistra chinensis Baker,with its structure determined and biological activity studied.The polysaccharides extracted from rhizome of Tupistra chinensis Baker were extracted by water extraction and alcohol precipitation.The 80%alcohol precipitation part TCP-C was purified by Sevage method.The polysaccharide TCP-C1-1 was purified from TCP-C by DEAE-52 cellulose ion exchange resin and dextran gel Sephadex G-200.The structure of polysaccharide TCP-C1-1 was analyzed by high effective gel chromatography,TLC thin layer chromatography,IR and 1H-NMR spectroscopy.The results showed that the TCP-C1-1 polysaccharide isolated and purified from the Tupistra chinensis Baker,was a homogeneous polysaccharide composed of fructoseβ-(2→1)connection with a relative molecular weight of 1.52×10^(4).TCP-C1-1 exhibited good biological activity.Its free radical scavenging rate was able to reach 63.3%at the concentration of 2 mg·mL^(-1) and its inhibition rate ofα-glycosidase was able to reach 78.6%at the concentration of 6.5 mg·mL^(-1).Besides,the proliferation of mouse monocyte macrophage RAW 264.7(p<0.05)was significantly promoted while the NO release induced by LPS was inhibited at the concentration of 1.5μg·mL^(-1).TCP-C1-1 is a natural inulin fructan isolated from Tupistra chinensis Baker for the first time,which provides a certain reference for the preliminary study and the development of polysaccharide components of Tupistra chinensis Baker.
