摘  要:为了明确高糖和益生菌复合饮食对人体口腔菌群的影响,研究分别考察了含有植物乳杆菌K25和鼠李糖乳杆菌GG的冰淇淋对12位受试者口腔菌群的影响。首先检测了益生菌冰淇淋对口腔主要病原菌Streptococcus mutans的抑制作用,与普通冰淇淋相比,K25和LGG冰淇淋使Streptococcus mutans活性下降了22%和38%(P<0.05),生物膜形成下降5.20%和10.35%(P<0.05),表面黏附率下降了18.10%和26.10%(P<0.05)。检测受试者食用冰淇淋后的口腔菌群发现,冰淇淋会影响口腔菌群多样性,其中普通冰淇淋组>K25冰淇淋组>未食用冰淇淋组>LGG冰淇淋组,普通冰淇淋口腔菌群α多样性增加了3.0%,K25组多样性增加了2.3%,但LGG组降低了7.8%,Beta多样性结果也显示LGG冰淇淋组的口腔菌群结构与未食用冰淇淋组相近。基于112个关键属的冗余分析显示,K25冰淇淋通过影响低丰度菌属增加口腔菌群多样性,而LGG冰淇淋主要通过影响口腔核心菌群(Veillonella和Streptococcus)的丰度来调节口腔菌群。总的来看,含有益生菌K25和LGG的冰淇淋表现出了定向调节口腔菌群能力,对龋齿和牙周炎相关病原菌群(Streptococcus等)具有较强的抑制作用,其中LGG的作用更强。To clarify the effects of probiotic ice cream on human oral microbiota,oral microbiota dynamics in 12 healthy persons after ingestion of ice cream fermented by Lactobacillus plantarum K25 and Lactobacillus rhamnosus GG were studied.Firstly,results of in vitro tests showed that K25 and LGG ice cream reduced activity of Streptococcus mutans by 22%and 38%compared with common ice cream(P<0.05).The biofilm formation of Streptococcus mutans was reduced by 5.20%and 10.35%(P<0.05),surface attachment rate reduced by 18.10%and 26.10%(P<0.05),respectively.The oral microbiota of 12 persons were significantly changed after eating ice cream,and the diversity of oral microbiota is ice cream group>K25 ice cream group>without-eating ice cream group>LGG ice cream group,in which diversity of ordinary ice cream group increased 3.0%,K25 group increased 2.3%,LGG group decreased 7.8%.Beta diversity analysis also showed that the oral microbiota composition of LGG ice cream group was similar to that of without-eating ice cream group.Based on the redundancy analysis of 112 key genus,we found that K25 ice cream increased the oral microbiota diversity by affecting low-abundance genus,while LGG ice cream moderated the oral microbiota by affecting the abundance of core oral microbiota(Veillonella and Streptococcus).Overall,we found that K25 and LGG ice cream showed the ability to target and regulate oral microbiota,with a strong inhibitory effect on dental caries and periodontitis related pathogens,and LGG has stronger effect.
