多棘海盘车多肽亚铁螯合物的制备研究

摘  要:以多棘海盘车为原料,采用木瓜蛋白酶酶解法制备得到多肽,通过单因素分析和正交实验优化多棘海盘车多肽与亚铁离子进行螯合反应的条件。结果表明:螯合反应的最佳条件为pH=3,多肽与亚铁离子质量比4∶1,反应温度65℃,反应时间50 min。在此条件下,Fe^(2+)配合率为46.98%,多肽亚铁螯合物得率为89.40%。红外光谱检测显示,Fe^(2+)与多肽中的氨基和羧基都有螯合反应。此次研究结果为多棘海盘车的加工利用开辟了一条新的途径,同时也为多肽亚铁螯合物工艺的发展提供理论基础。Asteriasamurensis was used as raw material tOproduce peptides by enzymolysis with papain.The conditions for the chelation reaction of peptides from Asteriasamurensis and ferrous chloride were optimized by single factor analysis and orthogonal test.The results showed that the optimal chelating condition was pH=3,mass ratiOof ferrous ions tOpolypeptide was 1∶4,chelating temperature was 65℃,and chelating time was 50 min.Under this condition,the Fe 2+complexation rate was 46.98%and the chelation rate was 89.40%.Infrared spectroscopy showed that Fe 2+has chelating reactions with aminOand carboxyl groups in the polypeptide.The results of this study have opened up a new approach for the processing and utilization of the Asteriasamurensis,and at the same time provide a theoretical basis for the development of the peptide ferrousiron chelate process.
