白玉豆中α-淀粉酶抑制剂的提取、纯化及其性质研究

摘　　要：α-淀粉酶抑制剂(α-AI)是一种常用于减肥和降低血糖的辅助药物,且其作为植物的抗虫基因在基因工程和药学应用广泛。从白玉豆中提取了α-AI并对其进行纯化,研究了其稳定性。使用CM纤维素柱和SephedexC-75凝胶柱对白玉豆α-AI粗品进行层析后,以HAc-NaAc洗脱液进行洗脱,收集蛋白峰后检测α-AI活性。结果表明,白玉豆中提取的白玉豆α-AI粗品可以通过层析得到α-AI。提取的α-AI耐热性能较好,在温度80℃以下,pH值3.0~10.0的范围内能保持比较高的生物活性,在pH值6.0~8.0范围内抑制活性最高。当料液比为1∶10g/mL,盐溶时间为8h时,经过CMII纤维素离子交换柱层析与SephadexG-75凝胶柱层析纯化后,得到的白玉豆提取的α-AI具有最高的生物活性。Alpha-amylase inhibitor(α-AI)is an adjuvant drug commonly used for losing weight and lowering blood glucose,and it is widely used in genetic engineering and pharmacology as an insect-resistant gene in plants.Theα-AI from Baiyu beans is extracted and purified,and its stability is studied.The crudeα-AI from Baiyu beans is chromatographed with a CM cellulose column and a SephedexC-75 gel column,then it is eluted with the HAc-NaAc eluent,and the protein peaks are collected and the activity ofα-AI is assayed.The result indicates that theα-AI can be obtained by chromatography from the crudeα-AI which is extracted from Baiyu beans.The obtainedα-AI has a better heat resistance,at a pH ranged from 3.0~10.0,it can maintain a relatively high biological activity below 80℃;and at a pH ranged from 6.0~8.0,the maximum inhibition activity is achieved.When the material-liquid ratio is 1:10g/mL and the salt dissolution time is 8h,theα-AI extracted from Baiyu beans can obtain the highest bioactivity after being purified by the CMII cellulose ion exchange column chromatography and the SephadexG-75 gel column chromatography.
