摘  要:以啤酒废酵母中提取的核糖核酸为原料,利用单因素试验和响应面优化试验优化了核酸酶P1催化水解核糖核酸的条件,并利用离子交换层析法对水解液中5′-核苷酸进行了初步分离鉴定。结果表明:最佳水解条件为核糖核酸质量浓度7.5 g/L、水解液初始pH 5、水解温度54℃、水解时间2.0 h。通过分离初步确定核糖核酸水解液中含有尿嘧啶核苷酸、鸟嘌呤核苷酸、胞嘧啶核苷酸和腺嘌呤核苷酸。In order to take the ribonucleic acid extracted from beer waste yeast as raw material, the conditions for the catalytic hydrolysis of ribonucleic acid by nuclease P1 were optimized by single factor test and response surface optimization test, and the 5′-nucleotides in the hydrolysate were preliminarily isolated and identified by ion exchange chromatography. The results showed that the optimal hydrolysis conditions were as follows: the mass concentration of ribonucleic acid 7.5 g/L, the initial pH of the hydrolysate 5, the hydrolysis temperature 54 ℃, and the hydrolysis time 2.0 h. Through separation, it was preliminarily determined that the hydrolysate of ribonucleic acid contained uracil nucleotide, guanine nucleotide, cytosine nucleotide and adenine nucleotide.
