摘 要:本研究以薏米蛋白为原料,采用碱性蛋白酶进行水解,在单因素试验基础上进行正交实验研究底物浓度、加酶量、反应时间对蛋白水解度(DH)的影响,经过优化得出制备薏米蛋白多肽(BPP)的最佳工艺为:反应温度55℃、反应pH9、底物浓度5%、加酶量1000U/g、反应时间4h。通过动物试验,采用自发性Ⅱ型糖尿病db/db小鼠模型,进行连续4周灌胃,测定小鼠血清指标,结果表明,与正常组相比,灌胃薏米蛋白多肽的中、高剂量组(200mg·kg^(-1)、400mg·kg^(-1))可以降低高血糖小鼠的空腹血糖、总胆固醇、甘油三酯含量(P<0.05),说明BPP可以有效调节高血糖小鼠的血糖和脂类的正常代谢。血清中抗氧化因子的测定结果表明灌胃BPP可以有效降低MDA水平,升高SOD和CAT水平,说明薏米蛋白多肽可以提高高血糖小鼠的抗氧化应激能力。In this experiment,barley protein was used as raw material,and alkaline protease was used for hydrolysis.On the basis of single factor experiment,an orthogonal experiment was carried out to study the influence of substrate concentration,enzyme amount and reaction time on the degree of proteolysis(DH).The optimal process for preparing barley protein polypeptide(BPP)was as follows:the reaction temperature was 55℃,the reaction pH was 9,the substrate concentration was 5%,the amount of enzyme added was 1000U/g,and the reaction time was 4h.The db/db mouse model of spontaneous type Ⅱ diabetes was used for 4 consecutive weeks of gavage,and the serum indices of the mice were measured.The results showed that compared with the normal group,the medium and high dose groups(200mg·kg^(-1),400mg·kg^(-1))could reduce the fasting blood glucose,total cholesterol,triglyceride content of hyperglycemic mice(P<0.05),indicating that BPP could effectively regulate blood glucose and normal metabolism of lipids in hyperglycemic mice.The measurement results of antioxidant factors in serum showed that intragastric administration of BPP could effectively reduce the level of MDA and increase the levels of SOD and CAT,indicating that the barley protein polypeptide could improve the anti-oxidative stress ability of hyperglycemic mice.
