摘 要:采用分光光度法分析葛根提取物与茶多酚有效成分的含量,对其复配后进行体内外辅助降血糖活性实验,通过α-葡萄糖苷酶活性的抑制效果验证体外降血糖活性,通过2型糖尿病小鼠的治疗效果验证体内降血糖活性。结果表明:葛根提取物中有效成分含量为(51.60±0.45)%,茶多酚中有效成分含量为(81.33±0.12)%,两者单独抑制时,半抑制浓度分别为1.000μg/mL与0.1159μg/mL,最佳的复配质量比为2∶1,复配时两者的半抑制浓度分别为0.1331μg/mL与0.06655μg/mL,联合指数为0.7073,小于0.9,具有良好的协同效果。体内实验中,采用400 mg/kg葛根提取物与200 mg/kg茶多酚对小鼠进行给药,其空腹血糖下降和血糖下面积降低效果显著;此外,该剂量给药对小鼠的胰岛素抵抗及胰岛素分泌指数均有一定改善作用。综上所述,葛根提取物与茶多酚复配后在体外α-葡萄糖苷酶的抑制效果与对高血糖小鼠的治疗效果均有协同效应。Active ingredients in a Pueraria extract and tea polyphenols were analyzed by spectrophotometry,and the auxiliary hypoglycemic activity of these compounds was tested in vitro(via inhibitory effect of theα-glucosidase activity)and in vivo(via therapeutic effect on type 2 diabetic mice).The results showed that the active ingredient content in the Pueraria extract was(51.60±0.45)%and(81.33±0.12)%in tea polyphenols.Their individual semi-inhibitory concentration was 1.000μg/mL and 0.1159μg/mL,respectively.The optimal combination mass ratio was 2∶1,and their semi-inhibitory concentration was 0.1331μg/mL and 0.06655μg/mL,respectively,when combined.Their combination index was 0.7073,well under 0.9,thus indicating a good synergistic effect.During the in vivo experiments,400 mg/kg Pueraria extract and 200 mg/kg tea polyphenols were administered to mice,and we observed that both fasting blood glucose and area under curve of glucose(AUC)decreased significantly.Furthermore,the insulin resistance and insulin secretion indexes of these mice improved upon administration of treatment doses.Moreover,the combination of the Pueraria extract and tea polyphenols showed a synergic effect onα-glucosidase inhibition in vitro and a synergic effect on hyperglycemic mice.
