摘 要:为了探讨褪黑素受体1A(MTNR1A)基因多态性对水牛繁殖性能的影响,试验以86头摩拉水牛经产牛为研究对象,采用PCR方法扩增其MTNR1A基因外显子Ⅱ序列,测序后分析该序列的多态性位点;记录86头摩拉水牛经产牛在2019年1月份—2020年12月份期间的怀孕和产犊情况,分析MTNR1A基因外显子Ⅱ的多态性与繁殖性能间的关系。结果表明:成功克隆到MTNR1A基因外显子Ⅱ,测序发现其第72位碱基处存在C>T单核苷酸多态性(SNP)位点,该位点有3种基因型,即CC、CT和TT基因型;C和T等位基因频率分别为0.47和0.53,CC、CT、TT基因型频率分别为0.37,0.30,0.33。CC和CT基因型摩拉水牛在2019年1月份和8—12月份时的怀孕数量均高于2019年2—7月份,TT基因型水牛在2019年2—7月份时的怀孕数量高于2019年1月份和8—12月份。摩拉水牛产犊季节主要集中在2020年1月份及9—12月份,经计算产犊数量占总产犊数的56.98%;2020年2—8月份是摩拉水牛产犊的淡季,经计算产犊数量仅占全年产犊总数的43.02%。CC和CT基因型的摩拉水牛产犊时间主要集中在2020年1月份和8—12月份,TT基因型水牛的产犊时间则变化不大。说明MTNR1A基因外显子Ⅱ第72位碱基处存在C>T SNP位点,可以作为一种基因标记,加速水牛良种选育进程。In order to investigate the effects of melatonin receptor 1 A(MTNR1 A) gene polymorphism on the reproductive performance of buffaloes, a total of 86 Murrah buffaloes were selected as subjects. The second exon sequence of MTNR1 A gene was amplified by PCR, and the polymorphisms was analyzed after sequencing. The pregnancy and calving of 86 parturitional Murrah buffaloes from January 2019 to December 2020 were recorded to analyze the relationship between the polymorphism of the second exon of MTNR1 A gene and its reproductive performance. The result showed that the second exon of MTNR1 A gene was successfully cloned, and C>T single nucleotide polymorphism(SNP) was found at site 72. There were three genotypes in this site, namely CC, CT and TT genotype. The frequency of C and T alleles were 0.47 and 0.53, respectively.The frequencies of CC genotype, CT genotype and TT genotype were 0.37, 0.30 and 0.33, respectively. The number of pregnancies in CC and CT genotypes of Murrah buffaloes in January 2019 and from August 2019 to December 2019 was higher than that from February 2019 to July 2019. The number of pregnancy in TT genotype buffalo from February 2019 to July 2019 was higher than that in January 2019 and from August to December in 2019. The calving season of Murrah buffaloes mainly occurred in January 2020 and from July to December in 2020, and the number of calving accounted for 56.98% of the total calving. From February to July in 2020 was the off-season for calving of Murrah buffaloes, and the number of calving only accounts for 43.02% of the total calving. The calving time of CC and CT genotypes of Murrah buffaloes mainly concentrated in January 2020 and from August to December in 2020, while there was little change in the calving quantity of TT genotypes of Murrah buffalo.These results indicated that there was a C>T SNP on the second exon of MTNR1 A gene, and this polymorphism could be used as gene marker to improve the reproduction rate and speed up the process of buffalo breeding.
