摘 要:目的研究蔗果三糖及低聚果糖(fructooligosaccharides,FOS)对T细胞分泌干扰素(interferon-,IFN-γ^(+))的调节作用。方法从外周血单个核细胞分离培养T细胞。在培养基中添加5mg/mL的蔗果三糖或FOS并检测T细胞的诱导分化。平行设置空白对照。经过3d的原代培养或6d的原代及传代培养,采用流式细胞术检测IFN-γ^(+)T细胞的比例,酶联免疫吸附法测定细胞培养液的IFN-γ^(+)浓度。结果经过3d的原代培养,蔗果三糖组IFN-γ^(+)T细胞比例(47.8100%±4.8800%)显著高于FOS组(36.9600%±4.5800%,P=0.0002)或空白对照组(31.4000%±3.8900%,P=0.0001),蔗果三糖组细胞培养液的IFN-γ^(+)浓度(848.5100 ng/mL±269.0000ng/mL)也显著高于FOS组(357.8700ng/mL±87.3000ng/mL,P=0.0040)或空白对照组(283.5100ng/mL±76.0300ng/mL,P=0.0001);而且,经过6d的原代及传代培养,无论传代之后是否添加蔗果三糖与FOS,蔗果三糖组IFN-γ^(+)T细胞比例(32.0600%±5.2600%;31.7900%±4.8600%)始终显著高于FOS组(22.1300%±4.5400%,P=0.0078;21.8900%±4.4800%,P=0.0086)或空白对照组(18.6500%±2.6900%,P=0.0071;17.8300%±3.1300%,P=0.0074)。结论蔗果三糖有助于T细胞分化并促进T细胞分泌IFN-γ^(+)。ObjectiveTo explore the regulatory effects of kestose and fructooligosaccharides(FOS)on interferon-(IFN-)secretion of T cell.Methods T cell isolated from peripheral blood mononuclear cells was cultured and proliferated.5 mg/mL kestose or FOS were added to the culture media of T cell to test the induced differentiation.A blank control was set in parallel test.After 3 days of primary culture or 6 days of primary culture and subculture,the percentage of IFN-γ^(+)T cell was assessed by using flow cytometry and the concentration of IFN-in the culture media was determined with enzyme linked immunosorbent assay.ResultsAfter 3 days of primary culture,the percentage of IFN-γ^(+)T cell in kestose group(47.8100%±4.8800%)was significantly higher than that in FOS group(36.9600%±4.5800%,P=0.0002)or blank control(31.4000%±3.8900%,P=0.0001).Meanwhile,the concentration of IFN-in the culture media from kestose group(848.5100 ng/mL±269.0000 ng/mL)was also significantly higher than that from FOS group(357.8700 ng/mL±87.3000 ng/mL,P=0.0040)or blank control(283.5100 ng/mL±76.0300 ng/mL,P=0.0001).And,the percentage of IFN-γ^(+)T cell in kestose group(32.0600%±5.2600%;31.7900%±4.8600%)was always higher than that in FOS group(22.1300%±4.5400%,P=0.0078;21.8900%±4.4800%,P=0.0086)or blank control(18.6500%±2.6900%,P=0.0071;17.8300%±3.1300%,P=0.0074)after 6 days of primary culture and subculture,with or without kestose and FOS in the culture media after cell passage.Conclusion Kestose helps T cell to differentiate and promotes the secretion of IFN-γ from T cell.
