摘 要:文章以杜氏盐藻为实验材料,对杜氏盐藻的培养进行单因素实验,针对碳源、氮源、磷源、盐度、钙离子和镁离子的种类及含量对该藻种生长及β-胡萝卜素、蛋白质和总糖等营养物质积累的影响。确定了各因素影响之下的最佳培养条件,根据各因素的最佳浓度对培养基进行优化,并针对优化后的培养基进行盐藻的培养和营养成分测定。结果表明:优化后的培养基对盐藻生长有显著的促进作用,盐藻的细胞密度最高可达(4.00±0.17)×10^(6) cells/mL,色素含量(叶绿素a、叶绿素b和β-胡萝卜素)分别达到了(18.36±0.66)μg/mL、(6.08±0.27)μg/mL和(7.80±0.43)μg/mL;蛋白质积累达到(2.74±0.16)mg/mL;总糖积累量为(2.27%±0.08%),均显著高于原培养基的细胞密度及营养物质积累量。Dunaliella salina is a large and versatile organism with high yield of glycerin andβ-carotene,and has great potential as an excellent feedstuff and raw materials for chemical and pharmaceutical products.In this article,the single factor experiment was carried out on the culture of Dunaliella salina.The effects of the types and contents of carbon source,nitrogen source,phosphorus source,salinity,calcium ion and magnesium ion on the growth and accumulation of nutrients such asβ-carotene,protein and total sugar of algae species were studied.The optimal culture conditions under the influence of various factors were determined,and the medium was optimized according to the optimal concentration of each factor,and the culture and nutrient composition of Dunaliella salina were determined for the optimized medium.The results showed that:The optimized medium significantly promoted the growth of Dunaliella salina,and the cell density of salina reached(4.00±0.17)×10^(6) cells/mL.The pigment contents(chlorophyll A,chlorophyll B andβ-carotene)were(18.36±0.66)μg/mL,(6.08±0.27)μg/mL and(7.80±0.43)μg/mL,respectively.Protein accumulation reached(2.74±0.16)mg/mL.The total sugar accumulation was 2.27%±0.08%,which were significantly higher than the cell density and nutrient accumulation of the original medium.
