摘  要:目的 利用高分辨飞行时间液质联用技术对茯砖茶发酵前后儿茶素类成分的含量变化进行研究。方法 利用儿茶素类成分对照品的色谱、质谱数据,比对保留时间、一级和二级精确质量数,定性分析茯砖茶中儿茶素类成分;基于各成分峰面积与内标物峰面积的比值,对发酵前后茯砖茶中儿茶素类成分进行定量分析,研究发酵后茶叶中儿茶素类成分的含量变化。结果 定性鉴定出茯砖茶叶中7个儿茶素类成分,分别为表儿茶素没食子酸酯(Epicatechin-3-gallate,ECG),表没食子儿茶素(Epigallocatechin,EGC),没食子儿茶素(Gallocatechin,GC),表没食子儿茶素没食子酸酯(Epigallocatechin gallate,EGCG),儿茶素(Catechin,C),表儿茶素(Epicatechin,EC),没食子儿茶素没食子酸酯(Gallocatechin gallate,GCG);同时,定量数据结果表明,发酵后的7个儿茶素类含量显著减少。结论 茯砖茶发酵前后,儿茶素类成分含量变化显著,且均呈现减小趋势,可为其发酵工艺和质量标准研究提供参考依据。Objective To study the content variation of catechins in Fuzhuan Brick-Tea before and after fermentation by liquid chromatography quadrupole time of flight tandem mass spectrometry(LC-MS). Methods Catechins in Fuzhuan Brick-Tea were identified by comparing retention time,precursor ions and product ions of samples with standards. To investigate the variation of catechins in Fuzhuan Brick-Tea before and after fermentation,the quantitative analysis of catechins was performed based on its relative content,which was the ratio of peak area of each compound to corresponding standard. Results Through the qualitative analysis,7 compounds were identified,included epicatechin-3-gallate(ECG),epigallocatechin(EGC),gallocatechin(GC),epigallocatechin gallate(EGCG),catechin(C),epicatechin(EC),gallocatechin gallate(GCG),respectively. Meanwhile,the results of quantitative analysis showed that the contents of 7 catechins had significantly reduced after fermentation. Conclusion The content of catechins in Fuzhuan Brick-Tea changed significantly before and after fermentation,and showed a decreasing trend. It can provide reference for the study of fermentation technology and quality standard.
