摘  要:为研究黄牛骨肽(cattle bone peptide,CBP)的免疫调节活性,采用巨噬细胞RAW264.7和氢化可的松诱导的免疫低下小鼠进行考察。基于细胞水平,考察不同浓度CBP对RAW264.7细胞增殖及吞噬功能的影响。通过各组小鼠的体质量变化、免疫器官指数,外周血白细胞计数、单核/巨噬细胞吞噬能力考察CBP的非特异性免疫活性。利用迟发型免疫反应和血清溶血素水平测定CBP的特异性免疫活性。体外细胞实验结果显示,CBP可以促进RAW264.7细胞的增殖且增强其吞噬中性红能力。与免疫低下组小鼠相比,CBP饲喂组小鼠的体质量、相对脾脏指数、相对胸腺指数、白细胞相对数量、碳廓清能力、吞噬荧光微球能力、血清溶血素水平均有不同程度改善,且免疫调节活性与剂量呈正相关性。研究结果表明,CBP可以增强机体非特异性免疫和特异性免疫活性,为CBP在食品及医药领域的进一步应用提供理论依据。The immunomodulatory activity of cattle bone peptide(CBP)was investigated based on RAW264.7 macrophages and immunosuppressed mice induced by hydrocortisone.At the cellular level,the effects of different concentrations of CBP on the proliferation and phagocytosis of RAW264.7 cells were investigated.The nonspecific immune activity of CBP was investigated by weight change,immune organ index,peripheral blood white blood cell count,and the phagocytosis ability of monocytes macrophages.Delayed-type hypersensitivity and serum hemolysin levels were used to determine the specific immune activity of CBP.The results of in vitro cell experiments showed that CBP enhanced the proliferation and the phagocytic capability to neutral red of RAW264.7 cells.Compared to the immunosuppressed control group,most indexes of the mice treated with CBP improved to varying degrees,including the body weight,relative spleen index,relative thymus index,relative white blood cell number,carbon clearance ability,phagocytic ability of fluorescent microspheres,and serum hemolysin levels.Moreover,the immunomodulatory activity and dosage of CBP were positively correlated.The results show that CBP can enhance the body’s non-specific and specific immune activity,thereby providing a theoretical basis for further application of CBP in the field of food and medicine.
