摘 要:从江苏淮安清江浦区柳树湾树林获取土样为菌种来源,以羧甲基纤维素钠(carboxymethyl cellulose sodium,CMC-Na)为唯一碳源,通过富集培养、初筛、复筛获得3株产纤维素酶能力较强的菌株(Strain-1、Strain-2、Strain-3,S-1、S-2、S-3),其中S-2产纤维素酶能力最强,其滤纸酶(filter paper enzyme,FPA)酶活力可达0.21 U/mL。对S-2菌株进行紫外(ultraviolet,UV)诱变,筛选获得菌株SUV2-1。通过单因素试验和响应面试验设计,对该菌株进行发酵培养基优化,得到该菌株产纤维素酶培养基配方:秸秆粉11.63 g/L,蛋白胨2.33 g/L、吐温-802.71 mL/L。利用上述培养基在摇瓶发酵条件下得到的FPA酶活力为0.45 U/mL,与优化前菌株的FPA酶活力相比,提高了51.15%;与诱变前的出发菌株相比,酶活力提高了113.00%。Three filamentous fungi strains[Strain-1(S-1),Strain-2(S-2),and Strain-3(S-3)]were obtained using enrichment culture,and two rounds of screening using soil samples obtained from Liushuwan forest(Qingjiangpu District,Jiangsu Province,China),with sodium carboxymethylcellulose(CMC-Na)as the only carbon source.Among the three strains,S-2 had the strongest cellulase production ability with an FPA activity of 0.21 U/mL.To improve the cellulase production ability of S-2,the strain was subjected to ultraviolet mutagenesis resulting in the strain SUV2-1.Through single-factor experimentation and response surface experimental design,the fermentation medium for the strain was optimized.The optimal medium for cellulase enzyme production from SUV2-1 contained straw powder(11.63 g/L),peptones(2.33 g/L),and Tween-80(2.71 mL/L).FPA activity obtained using this medium under shaking flask fermentation conditions was 0.45 U/mL.This represented an FPA activity increase of 51.15%compared with the strain prior to fermentation optimization and a 113.00%increase compared with the original strain prior to mutagenesis.
