摘 要:研究核桃青皮乙醇提取物对人黑色素瘤细胞(A375)氧化应激损伤的保护作用及对DPPH、ABTS自由基的清除作用。以不同浓度H_(2)O_(2)作用不同时间来诱导A375细胞氧化损伤,采用CCK-8方法检测A375细胞活力。通过丙二醛(MDA)、超氧化物歧化酶(SOD)、总抗氧化能力(T-AOC)检测氧化作用。从DPPH、ABTS自由基的清除率检测抗氧化活性。H_(2)O_(2)在浓度400.00μmol/L,作用时间4 h为建立氧化应激模型条件。与模型组比较,核桃青皮乙醇提取物的乙酸乙酯萃取物和二氯甲烷萃取物可以降低细胞中MDA含量、提高SOD酶活力和总抗氧化能力,并明显改善细胞形态。核桃青皮乙醇提取物的乙酸乙酯萃取物和二氯甲烷萃取物对DPPH自由基的清除率分别为为86.67%和63.54%,ABTS自由基的清除率分别为为91.53%和74.02%。To investigate the protective effect of Juglans mandshurica ethanol extractson oxidative stress injury of human melanoma cells(A375)and its scavenging effect on DPPH and ABTS free radicals.Oxidative damage of A375 cells was induced by different concentrations of H_(2)O_(2) for different periods of time,and the viability of A375 cells was detected by CCK-8 method.Oxidation was detected by malondialdehyde(MDA),superoxide dismutase(SOD)and total antioxidant capacity(T-AOC).Antioxidant activity was detected from the scavenging rate of DPPH and ABTS free radicals.An oxidative stress model was established when the concentration of H_(2)O_(2) was 400.00μmol/L for 4 h.Compared with the model group,ethyl acetate extract and dichloromethane extract of the ethanol extract of walnut green bark could reduce the MDA content,increase the SOD enzyme activity and total antioxidant capacity in cells,and significantly improve the cell morphology.The scavenging rates of ethyl acetate extract and dichloromethane extract from the ethanol extract of Juglans mandshurica on DPPH radical were 86.67%and 63.54%,respectively,and the scavenging rates of ABTS radical were 91.53%and 74.02%,respectively.
