摘 要:目的探究宫颈癌组织内人类runt相关转录因子2(Runx2)与人类runt相关转录因子3(Runx3)基因的水平及其临床意义。方法前瞻性选取手术或活检获得的宫颈组织标本100例,其中宫颈癌组织47例、宫颈上皮内瘤变(CIN)Ⅰ组织22例、CINⅡ组织18例、正常宫颈组织13例。运用实时荧光定量聚合酶链式反应(PCR)法检测宫颈组织内的Runx2 mRNA和Runx3 mRNA水平。结果宫颈癌组织内的Runx2 mRNA和Runx3 mRNA水平[(0.009±0.001)、(0.008±0.001)]低于CINⅠ组织、CINⅡ组织、正常宫颈组织,CINⅡ组织内的上述两种基因水平[(0.020±0.002)、(0.019±0.003)]低于CINⅠ组织、正常宫颈组织,CINⅠ组织内的两种基因的水平[(0.025±0.004)、(0.027±0.004)]低于正常宫颈组织,有统计学差异(P<0.05)。对47例宫颈癌组织检测表明,不同年龄、不同癌症类型、是否为高危型HPV感染与Runx2阳性表达无关(P>0.05),而不同分化程度、是否有淋巴结转移、不同FIGO分期与Runx2阳性表达有关(P<0.05);不同年龄、不同癌症类型、是否为HPV感染、是否有淋巴结转移与Runx3阳性表达无关(P>0.05),而不同分化程度、不同FIGO分期与Runx3阳性表达有关(P<0.05)。结论Runx2和Runx3基因在正常宫颈组织与宫颈癌组织内呈现出不同表达水平,其检测结果可作为诊断宫颈癌的参考依据。Objective To explore the level and other clinical significance of human runt-related transcription factor 2(Runx2)and human runt-related transcription factor 3(Runx3)genes in cervical cancer tissues.Methods 100 cervical tissue specimens were obtained by surgery or biopsy,including 47 cervical cancer tissue,22 cervical intraepithelial neoplasia(CIN)I tissue,18 CIN tissue and 13 normal cervical tissue.Runx2 mRNA and Runx3 mRNA levels were measured by quantitative real-time polymerase chain reaction(PCR).Results The Runx2 mRNA and Runx3 mRNA levels within the cervical cancer tissues[(0.009±0.001)、(0.008±0.001)]were lower than the CINI tissue,CIN tissue,normal cervical tissue,The above 2 gene levels within the CIN tissues[(0.020±0.002)、(0.019±0.003)]were lower than the CINI tissues,normal cervical tissues,The levels of both genes within the CINI tissues,which were[(0.025±0.004)、(0.027±0.004)],were lower than those in normal cervical tissues,Statistical difference(P<0.05).Detection of 47 cervical cancer types,different age,high-risk HPV infection were not associated with the Runx2 positive expression(P>0.05),while different differentiation,different FIGO stages were associated with Runx2 positive expression(P<0.05);different age,HPV infection,lymph node metastasis were not associated with the positive Runx3 expression(P>0.05),and different FIGO stages were associated with Runx3 positive expression(P<0.05).Conclusion The Runx2 and Runx3 genes show different expression levels in normal cervical tissues,and their test results can be used as a reference for the cervical cancer diagnosis,and can be vigorously promoted in clinical practice.
