摘 要:目的:基于响应面法建立焦山楂类黑精的最佳超声提取方法,并探究其体外抗氧化活性。方法:分别考察了超声温度、超声时间、超声功率、提取溶剂乙醇比、料液比五个因素对超声提取焦山楂类黑精提取量的影响,在此基础上通过响应面法对工艺经行优化,得出最佳提取工艺。按得到的最佳工艺制得焦山楂类黑精样品,用超滤管将其分为三种分子量梯度的样品并测定其类黑精含量。后依次测定了三种样品体外抗氧化活性(包括DPPH自由基清除率、ABTS自由基清除率和羟基自由基清除率),并探究其与类黑精含量之间的关系。结果:实验得出的最佳焦山楂类黑精最佳提取工艺为提取温度25℃;超声功率100 W;超声时间88.50 min;溶剂乙醇比53.63%;料液比选择1∶17。在三种不同分子量的样品中,H-MPRs中焦山楂类黑精的含量最高。焦山楂类黑精主要为分子量大于50 kDa的物质并且其具有较强的自由基清除能力,体外抗氧化作用明显且与其浓度呈正相关。结论:本研究基于响应面法建立了一种超声提取焦山楂类黑精的方法,并且发现焦山楂类黑精具有显著的体外抗氧化作用。Objective:Response surface methodology was used to establish the optimal ultrasonic extraction method of from hawthorn.And to investigate its antioxidant activity in vitro.Methods:The effects of ultrasonic temperature,ultrasonic time,ultrasonic power,ethanol concentration and liquid material ratio on the extraction amount of melanoidins from Hawthorn were investigated.On this basis,response surface method was used to optimize the extraction process.And the optimal extraction process was obtained.The samples were divided into three molecular weight gradients(H-MPRs,M-MPRs and L-MPRs)by ultrafiltration tube and the content of the melanoidins from samples was determined.The antioxidant activities(DPPH free radical scavenging rate,ABTS free radical scavenging rate and hydroxyl free radical scavenging rate)of the three samples were determined successively.And the relationship between the antioxidant activities and the content of melanoidins was explored.Results:The optimal extraction process of the black essence of hawthorn was as follows extraction temperature 25℃;Ultrasonic power 100 W;Ultrasonic time 88.50 min;ethanol concentration 53.63%;liquid material Ratio 1∶17.Among the three samples with different molecular weight,the content of the melanoidins from hawthorn in H-MPRS was the highest.The melanoidins from hawthorn was mainly substances with molecular weight greater than 50 kDa.And it has strong free radical scavenging ability.In vitro antioxidant activity was significant and positively correlated with its concentration.Conclusion:In this study,a method of ultrasonic extraction of melanoidins from charred hawthorn was established based on response surface method.And the melanoidins from charred hawthorn had significant antioxidant effect in vitro.
