摘 要:紫花凤梨(Tillandsia cyanea)是一种空气凤梨,具有很高的观赏价值。由于其基因组信息缺乏,其分子生物学相关研究鲜有报道。为研究紫花凤梨叶绿体微卫星(cpSSR)特征,本研究分析了紫花凤梨叶绿体全基因组序列(157602 bp),使用MISA软件共识别出204个SSR位点,位点间平均分布距离为772.6 bp。其中cpSSR主要集中于大单拷贝区(LSC),占SSR位点总数的64.22%。重复序列中主要以单碱基重复序列为主,占SSR位点总数的62.75%,主要由A/T碱基组成。cpSSR长度集中在8~20 bp,占95.10%,有10个cpSSR长度大于20 bp,这些cpSSR位点具有多态性的潜能。本研究结果为紫花凤梨的cpSSR引物筛选和遗传多样性等研究提供信息。Tillandsia cyanea is an epiphytic bromeliad,which has a very high ornamental value.Owing to the lack of its genomic resource,the development and application of molecular biology of T.cyanea is still limited.The SSR makers of T.cyanea in chloroplast genome were searched and analyzed by MISA.A total of 204 cpSSR loci were identified in the 157602 bp chloroplast genome of T.cyanea.The average distance between the loci was 772.6 bp.Most of the cpSSR loci distributed in the large single-copy region(LSC).The most common repeat type of the cpSSR is mononucleotide repeat.They accounted for 62.75%.The cpSSR were mainly made out of A and T bases.The length of cpSSR were concentrated in the range of 8~20 bp,accounting for 95.10%.There are 10cpSSR of length greater than 20 bp.The 10 cpSSR had the potency of high polymorphism.The results of this study will provide information for the cpSSR primer screening and genetic diversity of T.cyanea.
