摘  要:目的:基于多元统计分析法对半夏不同炮制品中8种潜在毒性成分与肝毒性的相关性进行分析。方法:采用UPLC-MS/MS法测定生半夏、清半夏、姜半夏、法半夏不同炮制品中溶血磷脂酰胆碱、盐酸麻黄碱、葫芦巴碱、胆碱、原儿茶酸、原儿茶醛、尿黑酸和大黄酚8种潜在毒性成分的含量。将半夏及其炮制品水提液给予大鼠灌胃给药后,测定大鼠血清中肝功能相关指标(ALT、AST、TBA、TBIL、AKP、IL-18、IL-1β、NLRP3、TNF-α)的水平;对半夏及其炮制品中8种潜在毒性成分与肝毒性的相关性进行多元统计分析。结果:与生半夏相比,清半夏、姜半夏、法半夏中溶血磷脂酰胆碱、胆碱、葫芦巴碱、盐酸麻黄碱、原儿茶酸和大黄酚的含量均降低,姜半夏、清半夏中原儿茶醛的含量升高,姜半夏、清半夏、法半夏中尿黑酸含量升高;半夏及其炮制品对正常大鼠可造成不同程度的肝毒性,经统计分析表明,溶血磷脂酰胆碱、盐酸麻黄碱、胆碱、原儿茶酸、葫芦巴碱、大黄酚与大鼠肝毒性相关。结论:通过炮制降低半夏肝毒性的作用可能与降低清半夏、姜半夏、法半夏中溶血磷脂酰胆碱、胆碱、盐酸麻黄碱、原儿茶酸、葫芦巴碱、大黄酚的含量有关。Objective: To analyze the correlation between 8 potential toxic components in different processed products of Pinellia ternata and hepatotoxicity based on multivariate statistical analysis. Methods: The contents of lysophosphatidylcholine, ephedrine hydrochloride, trigonelline, choline, protocatechuic acid, protocatechualdehyde, urinary black acid and chrysophanol in different processed products of SBX, QBX, JBX and FBX were determined by UPLC-MS/MS. After oral administration of Pinellia ternata and its processed water extract to rats, the related indexes of liver function(ALT, AST, TBA, TBIL, AKP, IL-18, IL-1β, NLRP3, TNF-α) were detected. The correlation between liver toxicity and 8 potential toxic components in Pinellia ternata and its processed products was analyzed by multivariate statistics. Results: Compared with SBX, the contents of lysophosphatidylcholine, choline, trigonelline, ephedrine hydrochloride, protocatechuic acid, and rhein were decreased in QBX, JBX and FBX;the content of protocatechualdehyde was increased in JBX and QBX;the content of homogentisic acid was increased in JBX, QBX and FBX;Pinellia ternata and its processed products could cause varying degrees of hepatotoxicity to normal rats. Statistical analysis showed that lysophosphatidylcholine, ephedrine hydrochloride, choline protocatechuic acid, trigonelline and chrysophanol had stronger correlation with rat hepatotoxicity. Conclusion: The effect of processing to reduce the hepatotoxicity of Pinellia ternata may be related to the contents reduction of lysophosphatidylcholine, choline, ephedrine hydrochloride, protocatechuic acid, trigonelline and chrysophanol in QBX, JBX and FBX.
