PD-1、PD-L1在乳腺癌中的信号表达及对调节性B细胞的诱导意义

摘  要:目的探讨程序性死亡受体1(PD-1)与程序性死亡受体配体1(PD-L1)在乳腺癌中的信号表达及对调节性B细胞的诱导意义。方法选择2018年10月至2020年10月我院收治的60例乳腺癌患者作为观察组,同期60例乳腺良性疾病患者作为对照组,比较两组乳腺组织中的PD-1、PD-L1蛋白表达情况。比较观察组中不同临床特征乳腺癌患者的PD-1、PD-L1蛋白表达情况。将观察组患者的石蜡切片分为A组、B组、C组各20例,采用三种不同浓度s PD-L1蛋白对CD19^(+)B细胞进行诱导培养,比较三组样本的Bregs比例。结果观察组的PD-1与PD-L1蛋白阳性表达率分别为31.67%,43.33%,均高于对照组的8.33%与11.67%(P<0.05)。观察组中不同临床特征乳腺癌患者的PD-1、PD-L1蛋白阳性表达率比较,差异均无统计学意义(P>0.05)。C组的Bregs比例高于B组和A组(P<0.05)。结论乳腺癌组织中PD-1、PD-L1蛋白具有较高的阳性表达率,高表达的s PD-L1蛋白对调节性B细胞具有较为明显的诱导作用。Objective To explore the signal expressions of programmed death receptor 1(PD-1)and programmed death receptor ligand 1(PD-L1)in breast cancer and its significance in the induction of regulatory B cells.Methods 60 patients with breast cancer admitted to our hospital from October 2018 to October 2020 were selected as the observation group,and 60 patients with benign breast diseases at the same period were selected as the control group.The expressions of PD-1 and PD-L1 proteins in breast tissues of the two groups were compared.The expressions of PD-1 and PD-L1 proteins in breast cancer patients with different clinical characteristics in the observation group were compared.The paraffin sections of the observation group were divided into group A,B and C,with 20 cases in each group.CD19^(+)B cells were induced and cultured with three different concentrations of s PD-L1 protein,and the proportions of Bregs in the three groups were compared.Results The positive expression rates of PD-1 and PD-L1 protein in the observation group were 31.67% and 43.33% respectively,higher than 8.33% and 11.67% in the control group(P<0.05).In the observation group,no significant difference was found in the positive expression rates of PD-1 and PD-L1 proteins in breast cancer patients with different clinical characteristics(P>0.05).The proportion of Bregs in group C was higher than that in group B and group A(P<0.05).Conclusions PD-1 and PD-L1 proteins have high positive expression rates in breast cancer tissues,and the high expression of s PD-L1 protein has obvious induction effect on regulatory B cells.
