摘 要:土槿皮粗分和蒸馏水以料液比1∶10混合,进行超声辅助提取,制备土槿皮样品溶液,浓度以生药质量计算。通过L-多巴氧化法研究土槿皮提取物对酪氨酸酶的抑制作用,并进行酶动力学作用机制分析;采用DPPH法和ABTS~+法研究土槿皮提取物的体外抗氧化能力。结果表明,土槿皮提取物对酪氨酸酶具有抑制作用,半抑制率IC_(50)值为(9.8±0.38)mg·mL^(-1),属于混合型抑制。同时,具有良好的DPPH自由基和ABTS~+自由基的清除能力,IC_(50)分别为(0.111±0.003)mg·mL^(-1)和(0.058±0.005)mg·mL^(-1)。土槿皮提取物具有酪氨酸酶活性抑制作用和显著的抗氧化能力,并呈现浓度依赖性关系,其酪氨酸酶抑制作用和抗氧化性表现出高相关性。The solution of Cortex Pseudolaricis extract was prepared by ultrasonic extraction, where Cortex Pseudolaricis powders and distilled water were blend with the ratio of 1∶10. Its concentration was calculated based on the pristine cortex pseudolaricis powders. Inhibition effect of the extract upon tyrosinase activity was detected by dopa oxidation method. In order to further analyze the mechanism of action, enzyme kinetics experiment was carried out. DPPH and ABTS+methods were used to evaluate the antioxidant capacity of Cortex Psedolaricis. The results showed that the Cortex Pseudolarics extract had inhibiting effect upon tyrosinase, and its 50% inhibiting concentration(IC_(50)) was(9.8±0.38) mg·mL^(-1), the inhibition mechanism was determined to be mixed inhibition type. Meantime, it showed significant scavenging ability to DPPH free radical and ABTS+free radical, with IC_(50) of(0.111±0.003) mg·mL^(-1) and(0.058±0.005) mg·mL^(-1) respectively.The extract of Cortex Pseudolaricis had significant antioxidant capacity and inhibition effect on tyrosinase activity, which highly depended on the concentration of extract. The inhibiting effect on tyrosinase was highly related to the antioxidant capacity.
