摘 要:以体外构建表达β-葡萄糖醛酸苷酶(β-glucuridase,β-Glu)及其酶学活性测定实验为例,探讨了综合型、创新型、科研转化型生物科学综合实验的建设理念及虚拟仿真+微视频+线下实操的新型混合教学模式的应用。通过更新实验项目、设置开放性实验以及改进教学和考核模式等,实现将自然学科教学与现代网络技术的深度融合,大大提高了本科实验教学的质量。在整个实验教学中,学生能够系统地掌握引物设计、PCR体外扩增、蛋白质的提取、分离、纯化、鉴定及酶学活性分析等相关理论及实验技术。这不仅圆满地完成了教学任务,也为学生打下了坚实的科研基础。Construction and expression of β-glucuridase in vitro and its enzyme activity identification experiment are taken as an example to discuss the construction concept of “synthesization-innovation-scientific research transformation” and the application of the new mixed teaching mode of “virtual simulation+micro video+offline practice” in biological science comprehensive experiment. By updating experimental projects, setting up opening experiments and improving teaching and assessment modes, the reform and innovation of undergraduate experimental teaching are better promoted, and the deep integration of science and modern network technology is realized. In the whole experimental teaching, students can systematically master primer design, vitro PCR amplification, protein extraction, isolation, purification, identification and enzyme activity analysis and other related theories and experimental techniques. This not only successfully completes the teaching task, but also lays a solid foundation for students in scientific research.
