摘 要:为满足高温高浓度乙醇发酵对发酵菌株耐高乙醇浓度、耐高温的要求,本文以前期筛选出的高温马克斯克鲁维酵母菌株KM-1为出发菌株,对其进行常压室温等离子体(ARTP)诱变以提高其乙醇耐受性。细胞经诱变后,利用含乙醇平板初筛选出86株生长良好的突变菌株,再利用含4%(V/V)乙醇的5%YPD培养基进行了液体培养复筛,大部分突变菌株的生长能力优于出发菌株。选择5株生长优秀的突变菌株进行了摇瓶发酵,结果表明,在45℃、利用含3%(V/V)初始乙醇浓度的15%YPD培养基进行发酵的条件下,5株突变菌株的发酵结果均优于出发菌株,24h内的乙醇积累量比出发菌株提升37.9%~47.5%。本研究结果表明马克斯克鲁维酵母的耐乙醇能力可通过ARTP诱变进行提升,为高温高乙醇发酵提供了潜在菌株资源。In order to meet the strain requirement of high-temperature and high-ethanol fermentation, this study carried out atmospheric room temperature plasma(ARTP) mutagenesis to improve the ethanol tolerance of Kluyveromyces marxianus strain KM1. After the cells were mutated, 86mutant strains with good growth on the ethanol-containing plate were selected. The growth of these mutant strains was re-screened using a 5% YPD liquid medium containing 4%(V/V) ethanol and most of the mutant strains showed better growth capacity than the original strain. Five mutant strains were selected for flask fermentation assay using 15% YPD with 3%(V/V) initial ethanol at 45 ℃. The results showed that the ethanol production of the five mutant strains was better than that of the original strain, which was 37.9%~47.5% higher than that of the original strain. This study suggested that the ethanol tolerance of Kluyveromyces marxianus could be improved by ARTP mutagenesis and provided potential strain resources for high temperature and high ethanol fermentation.
