摘  要:肌细胞生成素(myogenin,MyoG)是骨骼肌转录因子MyoD家族的一员,在肌细胞的形成过程中起着中心调控作用。利用21窝不同日龄的二花脸猪(Susscrofa Erhua)共97头,根据日龄组分布分成1组(日龄阶段):49 d,2组:58~60 d,3组:61~70 d,4组:71~80 d,5组:81~90 d,6组:228 d;采用索氏脂肪抽提法和石蜡组织切片-图像分析法对其背最长肌及半腱肌的肌肉组织学特性进行了测定。根据MyoG基因PCR-RFLP多态性,对二花脸猪不同的MyoG基因型与相关性状即初生重、20日龄体重、断奶重及肌肉组织学特性的关系进行了分析。结果表明:(1)在二花脸猪不同的MyoG基因型间,初生重差异极显著(P<0.01),但20日龄体重、断奶重无显著差异;(2)背最长肌的干肉样中的粗脂肪百分含量、肌纤维面积、肌纤维面密度、脂肪面密度在1~6日龄组内不同的MyoG基因型间差异均不显著;平均直径在1~5组均表现为AB<BB,4组组内差异显著(P<0.05),6组表现为AB>BB;肌纤维密度在1~5组表现为AB>BB,4组组内差异显著(P<0.05),6组表现为AB<BB;(3)半腱肌中除6组肌纤维密度表现为AB<BB差异显著(P<0.05)外,其余性状表现为组内基因型间差异不显著。Ninety-seven Erhualian pig (Sus scrofa Erhua) from 21 litters were divided into 6 groups according to their day age distribution, group 1: 49 d, group 2: 58-60 d, group 3: 61-70 d, group 4: 71-80 d, group 5: 81-90 d, group 6: 228 d. The longissimus over the last second to third vertebra thoracica and the middle part of the semitendinosus were taken to study the muscular histological characteristics by tissue paraffin section-image analyzing method. The longissimus over the second to fifth vetebra lumbalis were taken for measurement of intramuscular fat (IMF) content by Soxhlet's method. The polymorphisms of myogenin(MyoG) gene were detected by PCR-RFLP. Based on MyoG genetic variation detection, the effect of MyoG genotype on birth weight, 20-day weight, weaning weight and muscular histological characteristics were analyzed. The results of Dancans analysis indicated: (1) Highly significant variance was found on birth weight (P<0.01) between different MyoG genotypes, but no significant variance existed on 20-day weight and weaning weight. (2) In longissimus, no significant variance was found on IMF content in dried matter, muscle fiber area, muscle fiber area percentage and fat area percentage in all groups between different genotypes. The average muscle fiber diameter of AB genotype animals was smaller than that of BB genotype animal in group 1, 2, 3, 4 and 5, but group 4 presented significant variance (P <0.05) and group 6 presented as AB>BB. Consequently, muscle fiber density present as AB>BB, only group 4 significantly, group 6 presented as AB<BB. (3) In semitendinosus, no significant variance were found on all muscular characteristics except the muscle fiber density presented significantly in group 6 as AB<BB (P <0.05).
