摘  要:目的旨在观察基底膜蛋白多糖(perlecan)对小鼠肌卫星细胞中肌肉生长抑制素(myostatin)和胰岛素样生长因子-1(IGF-1,也被称为促生长因子)等肌肉萎缩相关因子信号表达的影响。方法应用单一肌纤维卫星细胞分离培养技术,分离出高纯度肌肉卫星细胞,通过添加perlecan因子,利用蛋白免疫印迹法(Western blot)、定量RT-PCR等技术方法观察Perlecan对Myostatin和IGF-1信号表达的影响。结果通过添加perlecan因子能促使肌肉卫星细胞中Myostatin含量增加,以及下游信号通路smad磷酸化过表达;促使IGF-1含量减少,以及下游信号通路Akt磷酸化表达降低(P<0.05)。结论Perlecan可通过调节肌肉萎缩相关因子Myostatin和IGF-1表达,对肌肉萎缩发挥调控作用。Objective To investigate the effect of perlecan which is exist in neuromuscular junction as one of extracellular matrix molecules,on the signal expression of muscle atrophy-related molecule that Myostatin and IGF-1of the rabbits,pharyngeal muscle.The research may broaden that our horizon for exploring the treatment of dysphagia caused by pharyngeal muscle atrophy.Methods Select the mices’pharyngeal muscle and separate the high purity muscle satellite cells by using the internationally recognized single muscle fiber satellite cell isolation and culture techniques.After adding perlecan,we can cell-based observe the effect on the expression of molecular myostatin,which related to pharyngeal muscle atrophy,and insulin-like growth factor one(IGF-1)with the method of Western blot and quantitative RTPCR.Results Adding perlecan into muscle satellite cells effects the level of muscle atrophy factors.It may promote the increase of the level of Myostatin,and its downstream signaling pathway smad phosphorylation overexpression;and restrain the decrease of IGF-1level,and has decreased phosphorylation expression of its downstream signaling pathway Akt.Conclusion Perlecan could play an important regulatory role in muscle atrophy by regulating the signal pathway of muscle atrophy-related factor Myostatin and IGF-1.
