摘 要:以没食子酸为标准品,对Folin-Ciocalteu比色法测定澳洲坚果青皮中多酚含量的条件进行优化。结果表明,最优检测条件为:检测波长762 nm, Folin-Ciocalteu试剂加入量1 mL, 15%的Na_(2)CO_(3)溶液加入量3 mL,加蒸馏水定容至10 mL,显色温度40℃,显色时间60 min,多酚质量浓度在20~80μg/mL范围内与吸光度呈良好的线性关系(R^(2)=0.999 0)。该方法的平均回收率为100.65%,相对标准偏差为1.27%。该方法操作简便,重复性、稳定性好,精密度、回收率高,可用于澳洲坚果青皮多酚含量的测定。The Folin-Ciocalteu colorimetric method for the determination of polyphenols in the pericarp of Macadamia integrifolia was optimized with gallic acid as the standard substance. The results showed that the polyphenol concentration had a good linear relationship with the absorbance value in the range of 20-80 μg/mL(R^(2)=0.999 0). The best determination condition was that the detection wavelength was 762 nm, the amount of Folin-ciocalteu reagent was 1 mL, the amount of 15% Na_(2)CO_(3) solution was 3 mL and fixed volume to 10 mL with the distilled water, the color rendering temperature was 40 ℃,and the color developing time was 60 min. The average recovery of the method was 100.65% with a relative standard deviation of 1.27%. The method was simple, reproducible, stable, precise and had a high recovery rate, which could be used for the determination of polyphenol content in the pericarp of Macadamia integrifolia.
