摘  要:[目的]观察泄浊解毒方对溃疡性结肠炎(ulcerative colitis,UC)小鼠Notch1、Hath-1及MUC2蛋白表达的变化,探讨泄浊解毒方在UC小鼠治疗中的作用机制。[方法]选取37只SPF级C57 BL/6小鼠,雄性,随机抽取8只确定为空白组(N组),其余小鼠通过自由饮用DSS溶液建立UC模型。确认造模成功后,将剩余小鼠随机分为模型组(M组)、美沙拉嗪组(S组)、泄浊解毒方组(Z组),每组各8只。N组及M组小鼠给予0.9%氯化钠溶液灌胃,S组、Z组给予相应药物灌胃治疗,疗程14 d,治疗结束后处死全部小鼠,留取结肠组织标本。采用免疫组织化学法检测结肠组织MUC2蛋白表达,Western Blot检测小鼠肠上皮细胞Notch1及Hath-1蛋白表达。[结果](1)与N组相比,M组小鼠MUC2蛋白表达显著减弱,阳性细胞分布区域明显缩小(P<0.05);与M组相比,S组及Z组上皮细胞MUC2均表达增强,阳性细胞分布范围增大(P<0.05);S组与Z组相比较,差异无统计学意义(P>0.05)。(2)与M组相比,S组及Z组小鼠结肠组织MUC2及Notch1蛋白表达均有所升高(P<0.05)、Hath-1蛋白表达有所降低(P<0.05)。[结论]泄浊解毒方可通过增加UC模型小鼠结肠组织MUC2及Hath1蛋白表达,降低UC模型小鼠结肠组织Notch-1蛋白表达,促进肠道黏膜修复。[Objective]To observe the effect of Xiezhuojiedu recipe on the expression of Notch1,Hath-1 and MUC2 protein in mice with ulcerative colitis(UC),and to investigate the mechanism of Xiezhuojiedu recipe in mice with UC.[Methods]Total 37 male c57 bL/6 mice were involved,and 8 of which were used as blank group(N).The rest of the mice were induced into UC model by freely drinking of DSS solution and were randomly divided into three groups:model group(M),mesalazine group(S),and Xiezhuojiedu recipe group(Z),every group was treated with corresponding drug gavage.After 14 d,all mice were sacrificed.The pathological changes of colon tissues were observed by hematoxylin-eosin staining,the expression of MUC2 protein in colon tissues was observed by immunohistochemistry,and the expression of Notch1 and Hath-1 protein in intestinal epithelial cells was detected by Western blotting(wester blot).[Results](1)Compared with the blank group,the expression of MUC2 in colonic epithelial tissue the distribution area of positive cells of the model group was significantly decreased(P<0.05).Compared with the model group,the expression of MUC2 and the distribution range of positive cells in both the positive control group and the Xiezhuojiedu recipe group was increaseded after the medicine administration(P<0.05).There was no significant difference between the positive control group and the Xiezhuojiedu recipe group(P>0.05).(2)The expression of colon tissue MUC2 and Notch1 protein in S group and Z group was significantly higher than that in M group(P<0.05),and the expression of Hath-1 protein was lower than that in the group(P<0.05).[Conclusion]Xiezhuojiedu recipe can treat and prevent the occurrence of UC by increasing the expression of MUC2 and Notch1 protein,reducing the expression of Hath-1 protein,improving the general condition of mice,repairing intestinal mucosa.
