摘 要:目的探讨并比较以葛根提取物和藤茶粉为主要材料的两种天然产物配方对急性酒精中毒小鼠乙醇脱氢酶(ADH)和乙醛脱氢酶(ALDH)活性的影响。方法将18~22gSPF级雄性KM小鼠随机分为AL[A配方低剂量:470 mg/(kg·bw)]、AH[A配方高剂量:1400mg/(kg·bw)]、BL[B配方低剂量:400mg/(kg·bw)]、BH[B配方高剂量:1200mg/(kg·bw)]、空白对照组、模型对照组、阳性对照组[海王金樽:1500mg/(kg·bw)]共7组,A、B配方组和阳性对照组按照受试物对应剂量灌胃14d,模型对照组和空白对照组灌胃双蒸水[灌胃剂量:20ml/(kg·bw)],第15d除空白组外其余组灌胃52°白酒[灌胃剂量:22ml/(kg·bw)]后观察翻正反射消失时间;酒精灌胃后的1、2和3h分别进行眼眶取血检测血乙醇、乙醛含量变化趋势;取肝脏测定ADH和ALDH活性以及HE染色观察肝脏病理变化。结果AH、BL、BH组ADH活性高于模型组(P=0.002;P=0.003;P=0.003);A、B高低剂量ALDH活性高于模型组(P<0.001);AH组在2~3h血中乙醇和乙醛含量降低。结论A、B组方在醒酒功能上均有一定程度的效果,但综合评价AH组效果最好。Objective To explore and compare the effects of two natural product formulations with pueraria root extract and rattan tea powder as main materials on the activities of ethanol dehydrogenase(ADH)and acetaldehyde dehydrogenase(ALDH)in acute alcoholism mice.Methods The 20 g to 22 g SPF grade male KM mice were randomly divided into 7 groups:AL(low dose of formula A:470 mg/kg·bw),AH(high dose of formula A:1400 mg/kg·bw),BL(low dose of formula B:400 mg/kg·bw),BH(high dose of formula B:1200 mg/kg·bw),blank control group,model control group,and positive control group(Haiwang Jinzun;1500 mg/kg bw).Formula groups A,B,and positive control group were intragaxed for 14 days according to the corresponding dose of test material,and model control group and blank control group were intragaxed with double steaming water(intragaxed dose:20 ml/kg·bw).The disappearance time of righting reflex was observed after gavage of 52°white wine(gavage dose:22 ml/kg-bw)in other groups except blank group on day 15.Orbital blood samples were taken at 1,2,and 3 hours after alcohol intragastric administration to detect the trend of blood ethanol and acetaldehyde content.ADH and ALDH activities were determined in liver and pathological changes were observed by HE staining.Results The ADH activity in AH,BL,and BH groups was higher than that in model group(P=0.002,P=0.003,and P=0.003).The activity of high-dose ALDH A and B was higher than that of model group(P<0.001).The contents of ethanol and acetaldehyde in blood of AH group decreased at 2-3 hours.Conclusion Both A and B groups have some effects on sober-up function,but the comprehensive evaluation of AH group is the best.
