摘 要:以兰州百合多糖为原料,采用β-甘露聚糖酶进行酶解,通过单因素试验与L9(33)正交试验研究底物质量分数、酶添加量、酶解时间对还原糖生成量的影响,优化得出最佳工艺条件;同时以维C为对照,运用DPPH、ABTS自由基、羟自由基和超氧自由基试验来评价兰州百合多糖酶解产物的抗氧化活性。结果表明,兰州百合多糖的最佳酶解工艺为底物质量分数0.2%,酶添加量0.5%,酶解时间4 h,在此条件下还原糖的生成量为0.8062±0.0084 mg/mL。所得酶解产物对4种自由基的清除能力在试验浓度范围内均强于未经酶解兰州百合多糖的清除能力,且呈一定的质量浓度依赖关系。当质量浓度为4 mg/mL时,兰州百合多糖对羟自由基的清除率为69.16%,酶解后产物对羟自由基的清除率达92.18%,接近于阳性对照维C。酶解提高了兰州百合多糖的抗氧化活性。Using Lanzhou lily polysaccharide as raw material,it was hydrolyzed byβ-mannanase.The effects of substrate concentration,enzyme dosage and hydrolysis time on the yield of reducing sugar were studied by single factor test and L9(33)orthogonal test,and the optimal technological condition was obtained.Meanwhile,the DPPH,ABTS free radical,hydroxyl radical and superoxide radical experiments were conducted to evaluate the antioxidant activity of the enzymatic hydrolysis products of Lanzhou lily polysaccharide,with VC as the control.The results showed that the optimal enzymatic hydrolysis condition was substrate concentration 0.2%,enzyme dosage 0.5%,and enzymatic time 4 h.Under these conditions,the yield of reducing sugar was 0.8062±0.0084 mg/mL.The scavenging ability of the obtained enzymatic hydrolysis products to the four free radicals was stronger than that of un-enzymatic hydrolyzing Lanzhou lily polysaccharide in the experimental concentration range,and the scavenging ability was in a certain concentration-dependent manner.When the concentration was 4 mg/mL,the scavenging rate of un-enzymatic hydrolyzing Lanzhou lily polysaccharide to hydroxyl radicals was 69.16%,and the scavenging rate of products after enzymatic hydrolysis to hydroxyl radicals was 92.18%,which was close to the positive control VC.Enzymatic hydrolysis enhanced the antioxidant activity of Lanzhou lily polysaccharide.
