摘 要:通过测试DPPH和羟自由基的清除活性,研究废弃龙眼核和龙眼壳提取物的体外抗氧化活性及其与多酚和黄酮含量的相关性。结果显示,龙眼核中的总酚和黄酮含量均高于龙眼壳,龙眼核中总酚、黄酮含量分别为15.675 mg/g和41.104 mg/g,龙眼壳中总酚含量、黄酮含量分别为14.605 mg/g和22.436 mg/g。体外抗氧化实验显示,抗氧化能力:核>壳,均随着浓度增大而增大,当生药浓度均为0.05 g/mL时,龙眼核和龙眼壳对DPPH自由基的清除率达到98%和93%;生药浓度均为0.008 g/mL时龙眼核和龙眼壳对对羟自由基的清除率达到91%和80%。相关性研究表明龙眼核/壳提取物对DPPH和羟自由基清除能力与黄酮含量和多酚含量相关性极显著,其相关系数分别为0.907,0.852,0.852和0.981。研究表明,龙眼核核龙眼壳提取物均具有较强的体外抗氧化活性,且抗氧化活性与总酚含量和总黄酮含量之间有明显的相关性,表明龙眼核、壳抗氧化活性的物质基础为酚类或黄酮类成分。This study aimed to understand the correlation of the antioxidant activity with the content of phenolics compounds and flavonoids compounds in Dimocarpus longan Lour in vitro.The results indicated that the seeds had the higher content of phenolics compounds(15.675 mg/g)and flavonoids compounds(41.104 mg/g)than the Longan shell content of phenolics compounds(14.605 mg/g)and flavonoids compounds(22.436 mg/g).The antioxidant assays showed that longan seeds had stronger antioxidant ability on DPPH and Hydroxyl radical,at Crude drug concentration(0.05 g/mL),the scavenging rate of longan seeds and shells on DPPH was 98%and 93%respectively;at Crude drug concentration(0.008 g/mL),the scavenging rate of longan seeds and shells on Hydroxyl radical was 91%and 80%respectively.The results of correlation study showed that the content of flavonoids had a strong relationship with the DPPH radical(r=0.907),Hydroxyl radical(r=0.852)scavenging activity,while the content of phenolics also had a strong relationship with DPPH radical(r=0.852),Hydroxyl radical(r=0.981)scavenging activity This research indicated that there is a close relationship between the content of phenolics and flavonoids and the antioxidant activity of longan seeds and longan shells,and the material basis of antioxidant activity of Dimocarpus longan Lour may be phenolics and flavonoids.
