摘 要:本试验旨在探索一种新的柠檬酸钙制备工艺,能够使微生物在产酸的同时合成柠檬酸钙,为实现高效、便捷且环保地合成柠檬酸钙提供理论依据。试验初期筛选产酸优势菌,经鉴定确定其为曲霉属黑曲霉菌种。通过单因素试验初步得到柠檬酸钙合成最佳方案,分析得除温度外,其余因素对柠檬酸钙合成影响均显著(P<0.05)。以单因素试验结果为中心点,通过响应面法进一步验证该方案。结论:响应面试验结果模型拟合良好(失拟相=0.0725>0.05,不显著),但三个因素自身及两两交互作用均不显著(P>0.05)。最终确定在石粉0.5g/100mL,反应温度55℃,转速120 r/min,反应时间46 h时柠檬酸钙产量最好。To investigate a new preparation process of calcium citrate,which enables microorganisms to synthesize calcium citrate while producing citric acid,and provides theoretical basis for efficient,convenient and environment friendly synthesis of calcium citrate.The strains which had the potential to produce citric acid were screened and identified as Aspergillus Niger species.The optimum synthesis process of calcium citrate was preliminarily obtained by single factor experiment.The analysis showed that other factors except temperature had significant infiuence on the synthesis of calcium citrate(P<0.05).The response surface method(RSM)was used to verify the process criteria.Conclusion:The response surface test results fit the model well(misfitting phase=0.0725>0.05,not significant),but the three factors themselves and the interaction between the two factors were not significant(P<0.05).It was determined that the best calcium citrate yield could be obtained when the stone powder was 0.5g/100mL,the reaction temperature was 55℃,the rotation speed was 120r/min and the reaction time was 46 h.
