摘  要:关于海洋中产几丁质酶具有低温耐酸特性的研究报道极少,因此筛选出具有产低温耐酸特性的几丁质酶菌株具有现实意义。该研究从连云港连岛海域采集的海泥样品中,通过透明圈法筛选一株产几丁质酶的菌株CZW003,结合形态学特征观察、生理生化试验及16S rDNA扩增分析鉴定出菌株为还原亚硝酸盐寡养单胞菌(Stenotrophomonas nitritireducens)并作酶学特性研究。结果表明,该酶最适反应温度为35℃,且在5℃具有催化活性,在5~35℃之间酶活较为稳定;最适pH为6.0,在pH 5.0时酶活仍保持在95%以上,酸性环境下具有较好的稳定性;Mg^(2+)、Ca^(2+)、K^(+)对酶活具有促进作用,而Hg^(+)、Fe^(2+)、Ag^(+)及EDTA对酶活具有显著的抑制作用;用胶体几丁质为底物时酶活力最高。There are few research reports on the acid resistance of chitinase produced in the sea at low temperature, so it is of practical significance to screen the strains which could produce chitinase with acid resistance at low temperature. In this study, a chitinase-producing strain CZW003 is screened from marine mud samples collected from Lianyungang Liandao waters by transparent zone method. The strain is identified as Stenotrophomonas nitritireducens by the combination of morphological characteristics observation, physiological and biochemical tests and 16S rDNA amplification analysis, and the enzymatic characteristics are studied.The results show that the optimum reaction temperature of the enzyme is 35 ℃, and it has catalytic activity at 5 ℃. The enzyme activity is relatively stable at 5~35 ℃. The optimum pH is 6.0, the enzyme activity remaines above 95% at pH 5.0, and it has good stability in acidic environment. Mg^(2+), Ca^(2+), K^(+)can promote the enzyme activity, while Hg^(+), Fe^(2+), Ag^(+)and EDTA can significantly inhibit the enzyme activity. The enzyme activity is the highest when using colloidal chitin as the substrate.
