摘 要:本实验采用高脂血症大鼠模型,以姜黄素和/或有氧运动干预6 w,观察各组大鼠心脏组织形态,检测血脂水平、血清心肌肌钙蛋白I(cTnI)、心肌肿瘤坏死因子-α(TNF-α)以及沉默信息调节因子2同源蛋白1(SIRT1)/核因子-κB(NF-κB)通路相关蛋白质表达水平,研究姜黄素和/或有氧运动对高脂血症大鼠心肌损伤的保护作用。结果表明:姜黄素和/或有氧运动可显著改善血脂异常,降低血清cTnI和心肌TNF-α水平,上调心肌SIRT1蛋白质表达,下调心肌NF-κB和Ac-NF-κB蛋白质表达,减轻心肌组织受损程度,其中联合干预效果最佳。结果显示6周姜黄素和/或有氧运动干预可有效降低高脂血症大鼠心肌细胞炎症水平,缓解心肌损伤,其可能与调节SIRT1/NF-κB信号通路相关蛋白质表达、抑制炎症反应有关。To study the protective effects of curcumin and/or aerobic exercise on myocardial injury in hyperlipidemia rats,the hyperlipidemia rat model was used. Curcumin and/or aerobic exercise was used to intervene the rats for 6 weeks. The myocardial histomorphology of rats in each group was observed,and blood lipid levels,serum cardiac troponin I(cTnI)levels,myocardial tumor necrosis factor-α(TNF-α)levels and silent mating type information regulation 2 homolog 1(SIRT1)/nuclear factor-kappa B(NF-κB)signaling pathway-related protein expression levels were detected. The results showed that curcumin and/or aerobic exercise improved dyslipidemia significantly,reduced serum cTnI and myocardial TNF-α levels,upregulated myocardial SIRT1 protein expression,downregulated myocardial NF-κB and Ac-NF-κB protein expression,alleviated the extent of myocardial injury. Among them,the combined intervention was the most effective. The results suggested that 6 weeks of curcumin and/or aerobic exercise intervention reduced the inflammation level of cardiomyocytes effectively and alleviated myocardial injury in hyperlipidemic rats,and the mechanism of its protective effect might be related to the regulation of SIRT1/NF-κB signaling pathway-related protein expression and inhibition of inflammatory response.
