摘  要:毕赤酵母(Pichia pastoris)表达系统是目前生物工程中最流行和最标准的一种重组蛋白生产工具。毕赤酵母系统产蛋白质的优势主要在于重组蛋白在内质网中的适当折叠和胞外分泌功能。此外,由于毕赤酵母本身几乎不产生分泌蛋白,因此重组蛋白的纯化过程相对于其它系统而言较简便。虽然毕氏酵母表达系统的优势较突出,并适用于生产实践,但由于其目标蛋白产量不高,因此需要对构建好的表达菌株进行一定程度的过程优化。本文综述了毕赤酵母近些年的发展现状和提高其外源蛋白表达量的相关策略及产业化研究进展,并展望了此系统的产业化发展前景,为进一步的研究提供理论依据。Pichia pastoris expression system is one of the most popular and standard recombinant protein production tools in bioengineering.The advantages of the Pichia pastoris system for protein production are mainly due to the proper folding of recombinant proteins in the endoplasmic reticulum and their exocytosis function.In addition,because Pichia pastoris produces almost no secretory protein itself,the purification process of recombinant protein is relatively simple compared with other systems.Although the Pichia pastoris expression system has outstanding advantages and is suitable for production practice,the production of target protein is not high,so the constructed expression strain needs to be optimized to a certain extent.In this paper,the development status of Pichia pastoris in recent years,the related strategies to improve the expression of exogenous proteins and the research progress of industrialization are reviewed,and the industrial development prospect of this system is indicated,so as to provide theoretical basis for further research.
