摘  要:通过抗逆性、产酶能力等特性实验从鸡肠道中筛选到一株益生性能较好的枯草芽胞杆菌HS-33。HS-33能够很好的耐受人工模拟的胃肠道环境,SD大鼠毒性试验表明HS-33安全无毒害作用,同时具有较强的产淀粉酶、蛋白酶和纤维素酶的能力。对该菌株进行显微形态、生理生化试验和16S rDNA分子生物学鉴定,结果表明菌株HS-33属于枯草芽孢杆菌(Bacillus subtilis)。通过正交试验优化HS-33菌株发酵工艺,培养基最佳配方为:玉米粉2%,黄豆粉4%,pH为7.0,MgSO_(4)·7H_(2)O 0.03%,MnSO_(4)0.005 mmol/L,NaCl 0.6%;最佳发酵条件为:温度37℃,时间24 h,接种量5%,摇瓶转速220 r/min,装液量60 mL,在此条件下培养菌株菌体量可达到8.1×109CFU/mL。Through the detection of stress resistance,enzyme production and antibacterial activity,a Bacillus sp.strain HS-33 with good probiotic performance was screened from chicken intestine. HS-33 had good stability to the simulated gastrointestinal environment. The toxicity test on SD rats showed that HS-33 was safe,non-toxic. HS-33 also had strong antibacterial properties against pathogenic bacteria. In addition,HS-33 could produce protease,amylase and cellulase. Micromorphology,physiology and biochemistry tests and the comparison of 16S rDNA sequence showed that strain HS-33 belonged to Bacillus subtilis. The fermentation process of HS-33 strain was optimized by orthogonal test. The optimal formula of fermentation medium was as follows :corn flour 2%,soybean flour 4%,pH 7.0,MgSO_(4)· 7H_(2)O 0.03%,MnSO_(4)0.005 mmol/L,NaCl 0.6%. The optimal fermentation conditions were:fermentation at 37℃ for 24 h,inoculation amount 5%,rotation speed of shaking flask 220 r/min,and loading volume 60 mL. The final bacterial concentration could reach 8.1×10^(9) CFU/mL.
