摘  要:以宁夏枸杞子为原料,枸杞子多糖提取率为考察指标,优化复合酶辅助低共熔溶剂对多糖的提取工艺,筛选低共熔溶剂的最优组合;单因素试验考察低共熔溶剂的摩尔比、含水量、液固比、微波功率、辐射时间、复合酶添加量、酶解时间、酶解温度对多糖提取率的影响;星点-响应面法优化提取工艺,然后检测其抗氧化与抑菌活性。结果表明:氯化胆碱/乙二醇的提取率较好,单因素试验结果表明液固比、酶加入量、酶解温度、微波功率是影响多糖提取的主要因素,星点-响应面法优化得到的最佳提取工艺为:液固比52 g/mL、酶加入量为1.37%、微波功率550 W、酶解温度47℃、辐射时间6 min、酶解时间2.5 h,在此条件下,枸杞子多糖提取率可达6.45%,与预测值相对误差为1.74%;体外活性研究结果显示纯化后的多糖对DPPH自由基、羟基自由基有一定的清除能力,抑菌活性结果表明多糖可以抑制大肠杆菌、金黄色葡萄球菌、枯草芽孢杆菌的生长。综上所述,复合酶辅助低共熔溶剂提取枸杞子多糖可有效提高提取效率,且提取纯化后的多糖能够保持良好的抗氧化与抑菌活性。Taking Ningxia wolfberry as raw material and polysaccharide extraction rate of wolfberry as index,the extraction process of polysaccharide with deep eutectic solvent assisted by complex enzyme was optimized,the optimal combination of deep eutectic solvents was screened.The effects of molar ratio of deep eutectic solvent,water content,liquid-solid ratio,microwave power,radiation time,compound enzyme addition amount,enzymolysis time and enzymolysis temperature on polysaccharide extraction yield were investigated by single factor experiment.The extraction process was optimized by star-point response surface,and the antioxidant and antibacterial activities were detected.The results showed that the extraction rate of choline chloride/ethylene glycol was better.The single factor test results showed that the ratio of liquid to solid,the amount of enzyme added,enzymolysis temperature and microwave power were the main factors affecting the extraction of polysaccharides.The optimal extraction process obtained by star-point response surface method was as follows:liquid-solid ratio 52 g/mL,enzyme dosage 1.37%,microwave power 550 W,enzymatic hydrolysis temperature 47℃,radiation time 6 min,enzymatic hydrolysis time 2.5 h,under these conditions,the extraction yield of polysaccharide from wolfberry could reach 6.45%,and the relative error was 1.74%.The results of in vitro activity study showed that the purified polysaccharides had certain scavenging ability on DPPH free radical and hydroxyl free radical.The results of antibacterial activity showed that the polysaccharides could inhibit the growth of Escherichia coli,Staphylococcus aureus and Bacillus subtilis.In conclusion,the extraction efficiency of wolfberry polysaccharide could be improved effectively with deep eutectic solvent assisted by complex enzyme,and the extracted and purified polysaccharide can maintain good antioxidant and antibacterial activities.
