摘 要:该研究建立了高效液相色谱(HPLC)同时测定天龙泉-陶藏酒中10种酚类物质(葛根素、槲皮素、大豆苷、山奈酚、芦丁、异槲皮苷、儿茶素、表儿茶素、槲皮苷、山奈酚-3-O-芸香糖苷)和2种三萜类物质(罗汉果苷IV、罗汉果苷V)的方法。确定检测条件为:Waters-Atlantis■T_(3)色谱柱(4.6 mm×250 mm,5μm),流动相A为乙腈,流动相B为水溶液,梯度洗脱,流速1.0 mL/min,进样量5μL,柱温32℃,检测波长203 nm,外标法定量。结果表明,该方法能快速准确检出天龙泉-陶藏酒中12种目标成分,各组分在质量浓度2~128 mg/L范围内有良好的线性关系,相关系数R^(2)为0.993~0.999,检出限为0.018~0.210 mg/L,定量限为0.062~0.701 mg/L,平均加标回收率为89.6%~119.7%,精密度试验结果相对标准偏差(RSD)为1.11%~4.28%。该方法操作简便、精密度好,能满足对天龙泉-陶藏酒中活性组分精准控制的需求。A HPLC method was established for simultaneous determination of 10 phenolic components(puerarin,quercetin,daidzin,kaempferol,rutin,isoquercitrin,catechin,epicatechin,quercitrin,kaempferol-3-O-rutinoside)and 2 triterpenoids(mogroside IV and mogroside V)in Tianlongquan Taocang Baijiu.The detection conditions were determined:Waters Atlantis■T_(3) column(4.6 mm×250 mm,5μm),acetonitrile as mobile phase,water as mobile phase B,gradient elution,flow rate 1.0 ml/min,sample loading volume 5μl,column temperature 32℃,detection wavelength 203 nm,and quantified by external standard method.The results showed that 12 target ingredients in Tianlongquan Taocang Baijiu could be quickly and accurately detected by the HPLC method,each component had a good linearity within the mass concentration in the range of 2-128 mg/L,the correlation coefficient R^(2) was 0.993-0.999,the detection limit was 0.018-0.210 mg/L,the limit of quantification was 0.062-0.701 mg/L,the average recovery was 89.6%-119.7%,the relative standard deviations(RSD)of precision tests results was 1.11%-4.28%.The method was easy to operate and had good precision,which could meet the needs of precise control of active components in Tianlongquan Taocang Baijiu.
