摘 要:目的 优化生物酶辅助提取连翘挥发油的工艺,并对连翘挥发油的主要成分进行分析。方法 采用生物酶辅助水蒸气蒸馏法提取连翘挥发油,以挥发油得率为响应值,采用Box-Behnken响应面法优选最佳提取条件;对提取得到的连翘挥发油进行GC-MS分析;以DPPH和OH自由基的清除率为指标,比较两种方法所得连翘挥发油的体外抗氧化能力。结果 最佳提取条件为:加酶量3.59%,酶解温度46℃,酶解时间2 h。该条件下挥发油平均得率为2.65%,与传统水蒸气蒸馏法比较,提取率提高了7.72%,且等体积连翘挥发油的提取时间明显缩短。与传统的水蒸气蒸馏法比较,生物酶辅助提取能提高连翘挥发油中的β-蒎烯、α-蒎烯、4-松油醇、D-柠檬烯的含量;两种方法得到的连翘挥发油均表现出DPPH和OH自由基清除能力。结论 生物酶辅助水蒸气蒸馏提取能明显提高连翘挥发油的提取率,提升药材资源的利用率,且工艺稳定可行,可为连翘挥发油的开发利用提供理论依据。OBJECTIVE To optimize enzymatic extration of volatile oil from Forsythia suspensa(VOFS),and to analyze the main components of VOFS.METHODS The VOFS was extracted by enzyme-assisted steam distillation method.The extraction technology of VOFS was optimized by using response surface methodology, and the extraction efficiency of VOFS was the response value.VOFS extracted by enzyme assisted extraction and steam distillation were analyzed by GC-MS.The scavenging capabilities of DPPH and OH radical were taken as indexes to compare the antioxidant capacity of VOFS by the two extraction methods in vitro.RESULTS The optimal process was as follows: enzyme addition of 3.59%,enzymatic temperature of 46 ℃,and enzymatic time of 2 h, the extraction rate of VOFS was 2.65%,which was 7.72% higher than steam distillation.In addition, the extraction time of VOFS was shortened obviously.The contents of β-pinene, α-pinene, 4-terpineol and D-limonene in the VOFS extracted by enzyme-assisted extraction were higher than the volatile oil extracted by steam distillation.The volatile oil obtained by the two methods had the ability to scavenge DPPH and OH free radicals.CONCLUSION Enzyme-assisted steam distillation can significantly improve the extraction rate of VOFS,and the utilization rate of medicinal materials.The process is stable and feasible, which can be the theoretical basis for the development and utilization of VOFS.
