摘　　要：目的该实验旨在研究AFP和AFB1对人源性肝癌细胞HepG2和Bel-7404细胞增殖、迁移及侵袭的影响。方法选取未做任何处理的HepG2和Bel-7404细胞作为空白对照组(NC组),以AFP药物处理组即为AFP组,以AFB1药物处理组即为AFB1组,以联合用药组为AFP+AFB1组。采用CCK8试剂盒检测各组细胞增殖能力的变化,划痕实验检测各组细胞迁移能力的改变,Transwell实验检测各组细胞侵袭能力的变化。结果与空白组比较,自第4天开始,在HepG2细胞的增殖、迁移及侵袭实验中均表现为NC组<AFP组<AFB1组<AFP+AFB1组(P<0.05);而侵袭实验12 h后表现为NC组<AFP组<AFB1组<AFP+AFB1组(P<0.05);与空白组比较,第4天开始,在Bel-7404细胞的增殖、迁移及侵袭实验中均表现为NC组<AFB1组<AFP组<AFP+AFB1组(P<0.05)。而侵袭实验12 h后表现为NC组<AFB1组<AFP组<AFP+AFB1组(P<0.05)。结论AFP和AFB1单独或联合作用对HepG2和Bel-7404细胞增殖、迁移及侵袭均具有促进作用,且二者具有协同作用,联合作用更明显。Objective To investigate the effects of AFP and AFB1 on the proliferation,migration and invasion of human-derived hepatocellular carcinoma cells HepG2 and Bel-7404.Methods HepG2 and Bel-7404 cells without any treatment were selected as blank control group(NC group);those treated with AFP drug as AFP group;those treated with AFB1 as AFB1 group;and those with combined drugs as AFP+AFB1 group.CCK8 kit was used to detect the changes of cell proliferation capacity of each group;scratch assay was used to detect the changes of cell migration capacity of each group;Transwell assay was employed to detect the changes of cell invasion capacity of each group.Results In comparison with the blank group,the proliferation,migration and invasion of HepG2 cells were shown as NC group<AFP group<AFB1 group<AFP+AFB1 group(P<0.05)since the fourth day,while after 12 h of invasion experiment,the results showed that NC group<AFP group<AFB1 group<AFP+AFB1 group(P<0.05).In comparison with the blank group,the proliferation,migration and invasion of Bel-7404 cells in assays were shown as NC group<AFB1 group<AFP group<AFP+AFB1 group(P<0.05)since the fourth day,while after 12 h of invasion experiment,the results showed that NC group<AFB1 group<AFP group<AFP+AFB1 group(P<0.05).Conclusion AFP and AFB1 alone or in combination can promote the proliferation,migration and invasion of HepG2 and Bel-7404 cells,and the two have a synergistic effect and the combined effect is more obvious.
