摘  要:目的 探讨灰毡毛忍冬皂苷乙(MB)在体外对人肝癌细胞株Hepal-6的抑制作用及可能机制的研究。方法 不同浓度的MB(0、 100、 200、 400μmol/L)作用肝癌Hepal-6细胞后,CCK-8法检测肝癌细胞株Hepal-6的存活率;通过测定了Hepal-6细胞中ROS的含量、MDA水平以及SOD、GSH、GSH-px的活性,探讨不同浓度的MB对Hepal-6细胞氧化应激水平的影响;通过检测FADD、Caspase8、RIPK1、RIPK3和MLKL的mRNA表达水平来检测不同浓度的MB对Hepal-6细胞坏死性凋亡相关基因的影响。结果 MB对肝癌Hepal-6细胞的抑制作用呈明显的剂量依赖性。结论 MB对肝癌细胞Hepal-6抑制存活率作用明显;MB破坏Hepal-6细胞内氧化还原系统的动态平衡,最终启动凋亡程序。另外,MB对坏死性凋亡相关基因表达影响,进而诱导肝癌细胞的坏死性凋亡而实现。Objective To investigate the inhibitory effect of Lonicera macranthoides on Hepal-6 cell line in vitro and its possible mechanism.Methods After different concentrations of MB(0,100,200,400 μmol/L) acted on hepatoma Hepal-6 cells, CCK-8 method was used to detect the survival rate of hepatoma cell line Hepal-6.By measuring the content of ROS,MDA level and the activities of SOD,GSH and GSH-px in Hepal-6 cells, the effect of different concentrations of MB on oxidative stress level in Hepal-6 cells was discussed.The mRNA expression levels of FADD,Caspase8,RIPK1,RIPK3 and MLKL were detected to detect the effect of different concentrations of MB on Hepal-6 cell necrosis related genes.Results The inhibitory effect of MB on hepatoma Hepal-6 cells was dose dependent.Conclusion MB has obvious inhibitory effect on the survival rate of hepatoma cell Hepal-6.MB destroys the dynamic balance of the redox system in Hepal-6 cells and finally initiates the apoptosis process.In addition, MB affects the expression of necrosis related gene protein, thus inducing apoptosis and necrosis of liver cancer cells.
