摘 要:以脱氧胆酸钠为膜软化剂制备纳豆激酶传递体,在单因素试验基础上,通过响应面法优化纳豆激酶传递体的制备工艺,并对其形态、粒径及透皮性能进行评价。结果表明:最佳制备工艺为囊材质量比3.85∶1、药料质量比0.07∶1、超声时间6.5 min,在此条件下获得包封率平均为72.3%±1.3%;纳豆激酶传递体结构呈现类球形,平均粒径为(275.2±10.1)nm、Zeta电位为-38.7 mV,体外透皮符合Higuchi方程,具有一定的缓释作用。Nattokinase transfersomes was prepared with sodium deoxycholate as membrane softener.Based on the single factor test,the preparation process of nattokinase transfersomes was optimized by response surface methodology,and its morphology,particle size and transdermal properties were evaluated.The results showed that the optimal preparation technology were as follows:the mass ratio of material 3.85∶1,the mass ratio of drug and material 0.07∶1,the ultrasonic time 6.5 min.Under these conditions,the average encapsulation rate was 72.3%±1.3%.The structure of nattokinase transfersomes was spherical,the average particle size was(275.2±10.1)nm,and the Zeta potential was-38.7 mV.The in vitro transdermal permeability conformed to the Higuchi equation,and hada certain slow-release effect.
