摘  要:目的通过对羟自由基(·OH)诱导血管平滑肌细胞差异表达蛋白的系统生物信息学分析,探讨·OH调控血管平滑肌细胞的相关分子机制。方法对485个差异表达蛋白(337个上调和148个下调蛋白)进行功能和通路富集、亚细胞定位预测、蛋白-蛋白互作(PPI)和信号网络构建,以及基因-miRNA、转录因子-基因和蛋白-化学品互作分析。结果·OH诱导的血管平滑肌细胞差异表达蛋白显著富集在一系列细胞核和细胞质中及与蛋白质活动有关的通路上;在PPI网络中,连环蛋白β1、组蛋白脱乙酰酶1和部分核糖体、蛋白酶体蛋白可能是·OH调控的关键节点蛋白。此外,发掘出一批miRNA(例如mir-181a-5p)、转录因子(例如上游结合转录因子)和化学品(例如四氯二苯二氧芑),亦可能参与·OH对血管平滑肌细胞的调控。结论预测·OH可调控多种分子,影响血管平滑肌细胞功能。Objective To explore the molecular mechanisms involved in the regulation of vascular smooth muscle cells by hydroxyl radical(·OH)through a systematic bioinformatics analysis of·OH-induced differentially expressed proteins in vascular smooth muscle cells.Methods We performed functional and pathway enrichment,subcellular localization prediction,protein-protein interactions(PPI)and signalling network construction for 485 differentially expressed proteins(337 up-regulated and 148 down-regulated proteins).Gene-miRNA,transcription factor-gene and protein-chemical interaction were analyzed.Results·OH-induced differential expression of proteins in vascular smooth muscle cells was significantly enriched in a range of pathways in the nucleus and cytoplasm that were associated with protein activity.In the PPI network,connexinβ1,histone deacetylase 1,some ribosomal and proteasomal proteins may be key nodal proteins in the regulation of·OH.In addition,a number of miRNAs(e.g.mir-181a-5p),transcription factors(e.g.upstream binding transcription factors)and chemicals(e.g.tetrachlorodiphenyl dioxin)were identified that may also be involved in the regulation of vascular smooth muscle cells by·OH.Conclusion This experiment predicts that·OH regulates a variety of molecules that affect vascular smooth muscle cell function.
