摘 要:以静态吸附率和解吸率为指标,筛选出适合荷叶、罗布麻总黄酮纯化的最佳大孔树脂,再用其进行动态吸附考察;采用单因素试验和正交试验优化荷叶、罗布麻总黄酮的纯化工艺条件。结果表明:荷叶、罗布麻总黄酮纯化的最佳树脂均是AB-8型大孔树脂,荷叶总黄酮最佳纯化工艺条件为上样流速3.0 BV/h、上样质量浓度1.606 mg/mL、乙醇体积分数80%、上样量1.4 BV,在此条件下荷叶总黄酮含量为65.75%;罗布麻总黄酮最佳纯化工艺条件为上样流速1.5 BV/h、上样质量浓度2.394 mg/mL、乙醇体积分数80%、上样量1.5 BV,在此条件下罗布麻总黄酮含量为46.75%。Based on the static adsorption rate and desorption rate, the best macroporous resin suitable for the purification of total flavonoids from lotus leaf and Apocynum venetum L. was selected, and then its dynamic adsorption was investigated. The purification conditions of total flavonoids from lotus leaf and Apocynum venetum L. were optimized by single factor test and orthogonal test. The results showed that the best resin for the purification of total flavonoids from lotus leaf and Apocynum venetum L. were AB-8 macroporous resin. The best purification conditions of total flavonoids from lotus leaf were as follows: the flow rate of sample was 3.0 BV/h, the mass concentration of sample was 1.606 mg/mL, the volume fraction of ethanol was 80 %, and the volume of sample was 1.4 BV. Under these conditions, the total flavonoids content of lotus leaf was 65.75 %. The optimal purification conditions of total flavonoids from Apocynum venetum L. were as follows: the flow rate of sample was 1.5 BV/h, the mass concentration of sample was 2.394 mg/mL, the volume fraction of ethanol was 80 %, and the volume of sample was 1.5 BV. Under these conditions, the total flavonoids content of Apocynum venetum L. was 46.75 %.
