摘 要:试验旨在测定20种牡丹叶中总黄酮、总酚和芍药苷含量及其抗氧化活性。试验以20个不同品种牡丹叶为研究对象,采用紫外分光光度法测定总黄酮、总酚含量,高效液相色谱法测定芍药苷的含量,采用1,1-二苯基-2-三硝基苯肼(DPPH)自由基、2,2-联氨-双(3-乙基苯并噻唑啉-6-磺酸)二胺盐(ABTS)自由基清除试验评价其抗氧化能力;采用相关系数法对不同品种牡丹叶中总黄酮、总酚、芍药苷的含量与抗氧化活性相关性进行分析,主成分分析法进行聚类评价。结果显示,20种牡丹叶中总黄酮含量为20.71~54.03 mg/g,总酚含量为68.31~188.19 mg/g,芍药苷含量为20.50~45.10 mg/g。牡丹叶中总黄酮、总酚、芍药苷的含量与DPPH自由基、ABTS自由基清除率的半抑制浓度(EC_(50))均具有显著负相关性;主成分分析将牡丹叶聚为4类,按花株色系差异存在一定的聚类。研究表明,牡丹叶中总黄酮、总酚和芍药苷的含量可作为评价其抗氧化能力的指标性成分。The experiment was to determine the content of total flavonoids,total phenols and paeoniflorin in 20 peony leaves and their antioxidant activity.The total flavonoids and total phenols were determined by ultraviolet spectrophotometry and paeoniflorin was determined by high performance liquid chromatography.DPPH free radical and ABTS free radical scavenging test were used to evaluate its antioxidant capacity.Correlation coefficient method was used to analyze the correlation between the content of total flavonoids,total phenols and paeoniflorin in leaves of different varieties of peony and their antioxidant activity,and principal component analysis method was used to cluster evaluation.The results showed that the content of total flavonoids,total phenols and paeoniflorin in 20 kinds of peony leaves ranged from 20.71 to 54.03 mg/g,68.31 to 188.19 mg/g and 20.50 to 45.10 mg/g.The content of total flavonoids,total phenols and paeoniflorin in peony leaves were negatively correlated with EC_(50) of DPPH and ABTS free radical scavenging.The leaves of peony were grouped into four groups by principal component analysis,and there was some clustering according to color difference of flower plant.The study indicates that the content of total flavonoids,total phenols and paeoniflorin in peony leaves can be use as the index components to evaluate the antioxidant capacity of peony leaves.
