摘 要:以人参叶为原料,从中结晶制备Re单体,对残余的母液中皂苷进行酶转化制备Rg1,能提高Rg1的提取率.人参叶总皂苷的提取采用甲醇浸提法,采用大孔吸附树脂法分离人参三醇类皂苷,采用结晶法从人参三醇类皂苷中制备Re单体,利用Aspegillus sp.g39菌产酶催化转化母液(Re和Rg1混合物)中的Re生成Rg1,进而制备Rg1单体.人参茎和叶中的皂苷组成类似,人参叶占茎叶总质量的四分之一,总皂苷质量分数达13.8%(人参茎中总皂苷质量分数为0.66%),其中Re和Rg1的质量分数分别为32.3%和15.9%.结晶法制备得到Re单体纯度达98%以上,以人参叶原料的质量计算得率为1.9%.结晶后的母液中还含有47.8%的Re,采用酶法转化Re制备Rg1,酶反应产物经硅胶柱层析纯化法制备得到Rg1单体,纯度达95%,以人参叶原料的重量计算Rg1的得率为2.8%.该研究发现利用人参茎叶,采用醇提、结晶、酶转化的技术方法可大量制备人参皂苷Re和Rg1.其中使用的酶转化法专一性强,生成Rg1产物单一,制备方法简单,同时可大幅提高Rg1的制备得率.In this paper,ginseng leaves were used as raw materials,from which Re monomer was separated by crystallization,and Rg1 was prepared by enzymatic conversion of saponins in the residual mother liquor,which can improve the extraction rate of Rg1.Total saponins of ginseng leaves were extracted by methanol,and ginsenoside was separated by macroporous adsorption resin.Re monomer was prepared from ginseng triol saponins by crystallization,and Rg1 monomer was prepared by catalytic conversion of Re in mother liquor(mixture of Re and Rg1)by Aspegillussp.g39.The composition of saponins in ginseng stems and leaves was similar.Ginseng leaves accounted for a quarter of the total weight of stems and leaves,and the total saponin content reached 13.8%(the total saponin content in ginseng stems was 0.66%),in which the contents of Re and Rg1 were 32.3%and 15.9%respectively.The purity of Re monomer prepared by crystallization method was above 98%,and the yield was 1.9%based on the weight of ginseng leaves.The mother liquor after crystallization also contained 47.8%Re.Rg1 was prepared by enzymatic conversion of Re,and Rg1 monomer was prepared by silica gel column chromatography purification.The purity of Rg1 was 95%,and the yield of Rg1 was 2.8%based on the weight of ginseng leaves.In this study,it is found that ginsenoside Re and Rg1 can be prepared in large quantities by alcohol extraction,crystallization and enzyme transformation from ginseng stems and leaves.Among them,the enzyme conversion method is highly specific in producing single Rg1 product with simple operation,which can greatly improve the preparation yield of Rg1.
