摘  要:为了探究晋汾白猪背最长肌肌纤维类型和能量代谢的特点,试验采集6月龄晋汾白猪和大白猪背最长肌,采用HE染色检测肌纤维的直径和密度;通过qRT-PCR分别检测生肌调节因子、肌纤维类型标记基因、氧化磷酸化、糖酵解活性以及线粒体活性相关基因的表达模式;PCR扩增晋汾白猪UCP3基因的CDS序列,并进行信号肽预测分析,采用qRT-PCR和Western blot检测晋汾白猪和大白猪中UCP3基因的表达特性。结果表明,相较于大白猪,晋汾白猪背最长肌肌纤维直径较小,密度较大;晋汾白猪背最长肌中生肌调节因子MyoD和MyoG的表达量较低;Ⅰ型肌纤维标记基因Myh7的表达量较高,Ⅱ型肌纤维标记基因Myh2、Myh4和Myh1的表达量均较低;糖酵解活性相关基因LDHA和LDHB、氧化磷酸化相关基因UQCRC2和NDUFA9以及线粒体活性相关基因ATP5A1和PGC1α的表达量均较低;晋汾白猪UCP3基因的CDS序列共有927个碱基,且不存在信号肽;晋汾白猪背最长肌中UCP3基因表达量和蛋白含量均显著低于大白猪;UCP3在晋汾白猪背最长肌和皮下脂肪组织中的表达量较高,且90 d晋汾白猪背最长肌中的表达量明显高于0、180 d。综上,晋汾白猪背最长肌肌纤维直径较小,I型肌纤维含量较高,Ⅱa型和Ⅱb型肌纤维含量较低,糖酵解和氧化磷酸化的活性较低,且UCP3的表达量低于大白猪。The purpose of the experiment was to explore the fiber types and energy metabolism characteristics of the longissimus dorsi muscle of Jinfen White pigs.In this study,the longissimus dorsi muscle of 6 month old Jinfen White pigs and Large White pigs was collected,and the diameter and density of muscle fibers were detected by HE staining.The expression patterns of myogenic regulatory factor,muscle fiber marker gene,oxidative phosphorylation,glycolytic activity,and mitochondrial activity-related genes were detected by qRT-PCR.The CDS sequence of UCP3 gene of Jinfen White pigs was amplified by PCR,and the signal peptide was predicted and analyzed.The expression characteristics of UCP3 gene in Jinfen White pigs and Large White pigs were detected by qRT-PCR and Western blot.The results showed that compared with the Large White pigs,the muscle fiber diameter of the longissimus dorsi muscle of Jinfen White pigs was smaller and the density was larger.The expression of myogenic regulatory factors MyoD and MyoG in the longissimus dorsi muscle of Jinfen White pigs was low.The expression of type I muscle fiber marker gene Myh7 was high,while the expression of type II muscle fiber marker genes Myh2,Myh4,and Myh1 was low.The expression of the glycolytic-activity-related genes LDHA and LDHB,oxidativephosphorylation-related genes UQCRC2 and NDUFA9,and mitochondrial-activity-related genes ATP5A1 and PGC1αwas low.The CDS sequence of the UCP3 gene of Jinfen White pigs had a total of 927 bases,and there was no signal peptide.The expression and protein content of UCP3 gene in longissimus dorsi of Jinfen White pigs were significantly lower than those of large White pigs.The expression of UCP3 in the longissimus dorsi muscle and subcutaneous adipose tissue of Jinfen White pigs was higher,and the expression of UCP3 in the longissimus dorsi muscle of Jinfen White pigs at 90 days was significantly higher than that at 0,180 days.To sum up,the diameter of the muscle fiber of the longissimus dorsi muscle of Jinfen White pigs was smaller,the
