摘  要:[目的]分析菌种种龄和接种量的协同作用对纤维素酶活性的影响,为生产纤维素酶奠定理论基础。[方法]分别对单菌种(DQ1)和混合菌种(DQ1+DQ2)不同种龄和接种量进行发酵培养试验,测定各处理的纤维素酶活力,分析不同处理纤维素的变化趋势及差异。[结果]在培养过程中,不同种龄的纤维素酶活力都在第8天达到最大;单菌种接种量为1%和2%时种龄4的纤维素酶活力最高,分别达到19.57和20.50 IU/mL;混合菌种接种量为1%和2%时种龄1的纤维素酶活力最高,分别为21.21和20.82 IU/mL。在第8天时,相同菌种和接种量的不同种龄的纤维素酶活力有一定差异。[结论]采用该研究的单菌或混合菌生产纤维素酶时,最优接种条件是种龄4、接种量2%,可以获得较高的纤维素酶活力,并且生产成本较低。[Objective]To analyze the effect of the synergistic effect of strain seeding age and inoculation size on cellulase activity and lay a theoretical foundation for cellulase production.[Method]The fermentation experiments of single strain(DQ1)and mixed strain(DQ1+DQ2)with different seeding ages and inoculum sizes were carried out,and the change trend and difference of cellulase activities were analyzed.[Result]During the culture process,the cellulase activity of different ages reached its maximum on the 8th day.The cellulase activity of seeding age 4th reached the highest at 1%and 2%inoculation of single strain,they were 19.57 and 20.50 IU/mL,respectively.And the cellulase activity of seeding age 1st reached the highest at 1%and 2%inoculation of single strain,they were 21.21 and 20.82 IU/mL.At the 8th day,the cellulase activity of different seeding ages of the same strain and inoculation size had certain differences.[Conclusion]When using the single or mixed strain in this study to produce cellulase,the optimal inoculation conditions are seeding age 4th,inoculation size 2%,which can obtain higher cellulase activity and lower production cost.
