摘 要:目的研究定痫丸颗粒剂对海人藻酸(KA)诱导的小鼠癫痫行为的干预作用,通过群体神经元多通道在体电生理记录技术,探讨定痫丸颗粒剂对小鼠癫痫发作时海马脑区神经元放电模式的影响。方法取雄性野生型小鼠28只,随机分为生理盐水对照组、KA诱导癫痫模型组、定痫丸干预组及丙戊酸钠干预组,每组7只。定痫丸干预组给予定痫丸颗粒剂灌胃;生理盐水对照组及KA诱导癫痫模型组给予与定痫丸干预组同等剂量的生理盐水灌胃;丙戊酸钠干预组给予丙戊酸钠腹腔注射。每天1次,持续10 d。最后1次给药干预后,除生理盐水对照组,其余每组均给予海人藻酸腹腔注射以诱发小鼠癫痫发作并观察各组的行为变化。同步采集海马脑区群体神经元的放电活动。结果定痫丸干预组与KA诱导癫痫模型组比较,小鼠癫痫发作的潜伏期明显延长,癫痫持续状态总时程缩短,癫痫发作行为频率降低,并且癫痫发作时海马脑区神经元放电峰值下降。结论给予动物定痫丸颗粒剂提前干预,能有效改善小鼠癫痫症状,具有明显的抗惊厥效果。Objective To study the intervention effect of Dingxian Pill granules on epileptic behavior induced by kainic acid(KA)in mice,and to explore the effect of Dingxian Pill granules on neuron discharge frequency in the hippocampus of mice during epileptic seizure by multi-channel in vivo electrophysiological recording of population neurons.Methods 28 Male wild-type mices were randomly divided into four groups:saline control group,KA-induced epilepsy model group,Dingxian Pill intervention group and sodium valproate intervention group,with 7 mice in each group.Dingxian Pill intervention group was given Dingxian Pill granule by gavage.KA-induced epilepsy model group and saline control group were given the same dose of saline as the Dingxian Pill intervention group,once a day for 10 days.And the sodium valproate intervention group was given sodium valproate intraperitoneal injection,once a day for 10 days.After the last administration,except the saline control group,the other groups were injected intraperitoneally with kainic acid to induce seizures in mice.The behavioral changes of mice in each experimental group were recorded.The discharge frequency of group neurons in hippocampus was collected simultaneously.Results Compared with KA-induced epilepsy model group,the latency of epilepsy induced by kainic acid in Dingxian Pill intervention group was significantly prolonged,the duration of status epilepticus was shortened,and the frequency of epilepsy was reduced.The peak discharge of action potential of neurons in the hippocampal region of mice decreased during epilepsy.Conclusion Dingxian Pill can effectively improve epileptic symptoms in mice and has obvious anticonvulsant effect.
