摘 要:试验旨在研究使用双酶法制备饲料级小麦低聚肽。试验对比了食品级与饲料级蛋白酶水解谷朊粉的多肽含量,筛选出最佳的复合酶种类;在单因素试验基础上,利用正交试验优化酶解条件并测定可溶性小肽含量。结果显示,酶解工艺条件为谷朊粉添加量12%、pH值7.0、中性蛋白酶与碱性蛋白酶各添加3%、水解5 h。在此条件下,可溶性小肽含量84.34%。使用高效液相色谱测定其重均分子量为764 Da,分子量小于1 000 Da的小麦低聚肽占比87.19%。研究表明,使用饲料级蛋白酶酶解谷朊粉,小麦低聚肽含量显著提高,并降低成本,具有作为功能性饲料添加剂应用的潜力。The experiment was to study the preparation of forage grade wheat oligopeptide by dual enzymatic method.The peptide content of food grade protease and feed grade protease was compared,and the best compound enzyme was selected.On basis of single factor experiment,orthogonal experiment was used to optimize enzymolysis conditions and determine the content of soluble small peptides.The results showed that the enzyme hydrolysis condition was 12%of gluten,pH value of 7.0,neutral protease and alkaline protease added of 3%each,hydrolysis 5 h.Under these conditions,the content of soluble small peptide was 84.34%.The weight average molecular weight of wheat was 764 Da determined by HPLC,and 87.19%of wheat oligopeptide with molecular weight less than 1000 Da.The results show that the use of feed-grade protease enzyme can significantly increase the content of wheat oligopeptide and reduce the cost,which has the potential to be used as a functional feed additive.
